Effects of recombinant human lactoferrin on expressions of |pro-inflammatory cytokine IL-6 and IL-8 in Tca8113 cells

Abstract:Objective
To study the effects of recombinant human lactoferrin( rhLF ) on the expressions of pro-inflammatory cytokine IL-6 and IL-8   in Tca8113 cells in oral squamous cell carcinoma  and to explore the possible mechanism of inhibiting the growth of tumor cells.Methods The Tca8113 cells cultivated in vitro were treated with rhLF(50  and 100 mg•L-1).Enzyme linked immunosorbent assay and immunohistochemical method were used to detect the influence  of rhLF on the expressions of IL-6 and IL-8 proteins.Results The levels of IL-6 and IL-8 in supernatant were gradually  decreased with the increasing of the concentrations of rhLF.Compared with normal control group,the levels of IL-6 and IL-8  in  100 mg•L-1 rhLF group were significantly decreased (P<0.05).The results of immunohistochemical staining showed that the expressions of IL-6 and IL-8 were detected in Tca8113 cells.Compared with control group,the expressions of IL-6 and IL-8  in 50 mg•L-1rhLF group were significantly decreased,there was only weak expression or no expression in this group.Conclusion rhLF can specificially inhibit the generation of IL-6 and IL-8 in Tca8113 cells and induce the changes of inflammatory environment of tumor cells to inhibit the growth of tumor.
