Inhibitory effect of lactacystin on proliferationof glioma C6 cells in vivo

Abstract:Objective
To observe the effect of lactacystin (LAC) on the proliferation rate of C6 cells in vivo on the basis of inhibitory effect of LAC on C6 cells in vitro and  to discuss the possible mechanism of LAC  inhibiting the proliferation rate of C6 cells. Methods The  C6 cells were cultivated in vitro,and the cells in the logarithmic growth phase were selected and were inoculated in nude mice  subcutaneously.All mice were given LAC for 7 d.They were divided into model group,LAC 0.5 μg / 20 g group,LAC 1 μg / 20 g group,and the LAC 5 μg / 20 g group.There were three nude mice in each group.The general condition of mice (food,bathroom,coat color,activities,etc.) was observed;the tumor volumes at different time points were measured and calculated.The Bax,bcl-2 and caspase-3 mRNA expression levels were detected by RT-PCR method.Results Compared with model group, the tumor volumes  in LAC 0.5 μg / 20 g group,LAC 1 μg / 20 g Group,and  LAC 5 μg / 20 g group were significantly reduced(P<0.05);the Bax/bcl-2 and caspase-3 mRNA expression levels were increased significantly (P<0.05).There was not statistically significant between different doses of LAC in  tumor volume,level of Bax/bcl-2 mRNA and level of caspase-3 mRNA,respectively.Conclusion LAC in animals can significantly inhibit the proliferation rate of C6 cells and its mechanism may be that it increases   apoptosis of mitochondrial pathway in C6 cells to inhibit the proliferation.
