BXSB狼疮鼠骨髓粒-巨噬细胞集落形成单位异常与发病的关系

ARelationship between abnormality of colony-forming units of granulocytes and macrophages and pathogenesis in BXSB lupus mice

摘要: 目的:研究BXSB自发性狼疮鼠骨髓粒-巨噬细胞系的增殖情况与狼疮发病的关系。 方法:雄性BXSB小鼠按4、8、12、16及20周龄分组,每组6只,采用甲基纤维素半固体培养方法体外培养骨髓单个核细胞(BMMC),计数粒-巨噬细胞集落形成单位(CFU-GM)及小鼠外周血单核细胞(PBMC),并检测尿蛋白。相同周龄的C57BL小鼠作为正常对照组。 结果:12周龄雄性BXSB小鼠尿蛋白(+),20周龄时尿蛋白(),4、8周龄BXSB小鼠及各周龄C57BL小鼠尿蛋白(-)。8周龄雄性BXSB小鼠骨髓CFU-GM为(96.00±18.11)/2×105 BMMC,PBMC计数为(111.33±10.97)×106•L-1,均明显高于4周龄BXSB小鼠及正常C57BL小鼠(P<0.05),随着周龄增加,CFU-GM和PBMC进一步增多。 结论:BXSB小鼠粒-巨噬细胞系造血异常及外周血单核细胞增多参与了自身免疫疾病的发病过程。

Abstract: Objective To study the proliferation of colony-forming units of granulocytes and macrophages (CFU-GM) and relationship with pathogenesis in BXSB lupus mice. Methods The formation of CFU-GM of bone marrow mononuclear cells (BMMC) of 4, 8, 12, 16 and 20-week-old male BXSB mice cultured in vitro with methylcellulose semisolid culture system was observed, the peripheral blood monocytes (PBMC) were counted and the urinary protein was test at the same time. C57BL mice with same ages were used as normal controls. Results The urinary protein emerged since 12 weeks in male BXSB, but remained negative in 4, 8-week-old male BXSB and C57BL with any ages. The CFU-GM was (96.00±18.11)/2×105 BMMC, and peripheral blood monocytes reached (111.33±10.97)×106•L-1 in 8-week-old male BXSB mice, the counts were significantly higher compared with those of 4-week-old male BXSB mice and normal C57BL mice (P<0.05), and increased progressively in older BXSB mice. Conclusion The autoimmune disease of male BXSB mice may be caused by monocytosis which comes from the abnormal CFU-GM in the bone marrow.
