喉癌转移中黏附分子CD44v6蛋白表达和胞内环素α、β mRNA的表达

Expressions of CD44v6 protein and α- andβ-catenin mRNA in metastasis of laryngeal cancer

摘要: 目的:探讨转移相关因子CD44v6蛋白表达和胞内环素α和胞内环素β(α-catenin和β-catenin)的mRNA表达在喉癌转移中的作用。 方法:采用免疫组化ABC和RT-PCR方法,对64例喉癌组织和9例取自尸检的正常喉组织进行了CD44v6蛋白表达和胞内环素α、β mRNA表达的检测。 结果:CD44v6蛋白在正常对照组中均有表达(灰度值173.1±15.0),喉癌组织中9例表达缺失(灰度值>200),14例表达为弱阳性(灰度值186~200),CD44v6表达缺失或弱表达多发生在喉癌病理分化程度差或有淋巴结转移的标本中(P<0.05)。64例喉癌组织中,胞内环素α mRNA异常表达6例,其中4例表达阴性,2例缺失700 bp基因片段;胞内环素β mRNA异常表达10例,其中7例表达阴性,3例缺失350 bp基因片段。胞内环素α mRNA和胞内环素β mRNA表达异常与喉癌临床分期及淋巴结转移明显正相关(P<0.05)。 结论: CD44v6蛋白表达下调或缺失及胞内环素α、β mRNA表达异常可能在喉癌淋巴结转移过程中起一定的作用。

Abstract: Objective To explore the roles of abnormal protein expression of cluster of differentiation (CD44v6) and mRNA expressions of α- and β-catenin in metastasis of laryngeal cancer. Methods CD44v6 protein expressions and mRNA expressions of α- and β-catenin in 64 cases of laryngeal cancer and 9 cases of normal laryngeal tissues were detected with the methods of immunohistochemistry and RT-PCR. Results Positive protein expressions of CD44v6(Gray level:173.1±15.0)were observed in normal controls,and negative expressions(9 cases,Gray level:>200)or low expressions(14 caces,Gray level:186-200)of CD44v6 were found in the laryngeal cancer tissues. Abnormal expression of CD44v6 protein was correlated with histological grades and lymph node metastasis in laryngeal cancer. Abnormal expressions of α- and β-catenin mRNA were detected in 6 samples (negative:4 cases; deletion of 700 bp gene fragment:2 cases) and 10 samples (negative:7 cases; deletion of 350 bp gene fragment:3 cases) with laryngeal cancer, respectively. Abnormal expressions of α-catenin and β-catenin mRNA were apparently associated with clinical stages and lymph node metastasis in laryngeal cancer (P<0.05). Conclusion Both negative or low expressions of CD44v6 and abnormal mRNA expressions of α-catenin and β-catenin can play a role in the lymph node metastasis of laryngeal cancer.
