Effects of PEDF on proliferation,invasion and apoptosis of lung squamous cell carcinoma SK-MES-1 cells

Abstract:

Objective To explore the effects of pigment epithelium derived factor(PEDF) on the proliferation, apoptosis and invasion of lung squamous cell carcinoma SK-MES-1 cells, and to interpret the relationships between PEDF and the carcinogenesis and progression of lung squamous cell carcinoma. Methods The SK-MES-1 cells were primarily cultured.The cells without PEDF were usded as negative control group, the cells in PEDF intervention groups were treated with 180, 360, and 720 nmol·L-1 PEDF.The inhibitory rates of proliferation of SK-MES-1 cells were observed by MTT assay, the number of SK-MES-1 cells penetrating the membrane in negative control group and 720 nmol·L-1 PEDF group were determined by cell invasion experiment, the morphology of SK-MES-1 cells was observed by inverted microscope, and the apoptotic rates were detected by flow cytometry. Results The inhibitory rates of proliferation of SK-MES-1 cells in PEDF intervention groups were higer than that in negative control group(P<0.01), and there were statistics differences among the PEDF groups(P<0.05).The cells in negative control group grew well, but the morphological changes of the cells in intervention groups were obvious.The number of the cells penetrating the membrane in 720 nmol·L-1 PEDF intervention group (31.6±15.8) was lower than that in negative control group (75.4±19.3)(P<0.01).The apoptotic rates of SK-MES-1 cells were increased with the increasing of PEDF doses, and there were significant differences between different doses of PEDF intervention groups and negative control group (P<0.05). Conclusion PEDF inhibits the proliferation or invasion and promotes the apoptosis of SK-MES-1 cells, and play an effect of inhibiting the growth and metastasis of lung squamous cell carcinoma.PEDF may be a potent factor for the prognosis and treatment of lung squamous cell carcinoma.
