Inhibitory effect of wild chrysanthemum extract on activity of trypsin-like protease produced by Porphyromonas gingivalis

Abstract:

Objective To investigate the effect of wild chrysanthemum extract on the activity of trypsin-like protease (TLP) produced by Porphyromonas gingivalis(P.g),and try to provide the evidence for the wild chrysanthemum extract in the treatment of periodontal disease. Methods The brain heart infusion broth(BHI),and low,medium and high concentrations(0.1563,0.3125,and 0.6250 g·L-1) of diluted wild chrysanthemum extract and turbidity of P.g bacteria suspension were prepared.Low,medium,and high concentrations of wide chrysanthemum extract groups,positive control group (without wide chrysanthemum extract),negative control group (with low concentration of wild chrysanthemum extract and without P.g) were set up.The Ep tube with solution in various groups were cultured under 37℃ for 48 h.Then the TLP activity in the supernatant fluid of P.g and protein levels were detected.The specific activity of TLP was calculated. Results Compared with positive control group,the TLP activities in supernatant fluid in medium and high concentrations of wild chrysanthemum extract groups were decreased(P<0.05);compared with low concentration of wild chrysanthemum extract group,the TLP activities in supernatant fluid in medium and high concentrations of wild chrysanthemum extract groups was decreased(P<0.05);compared with medium concentration of wild chrysanthemum extract group,the TLP activities in supernatant fluid in high concentration of wild chrysanthemum extract group was decreased(P<0.05).There were no significant differences of the protein levels and TLP specific activities in supernatant fluid in various groups(P>0.05). Conclusion The wild chrysanthemum extract may inhibit the TLP activity.
