Therapeutic effect of α-2b interferon on inflammation of liver tissue and liver fibrosis in hepatitis B model ducks

Abstract:

Objective: To establish the duck models of hepatitis B by duck hepatitis B virus(DHBV) serum injection method,and to explore the effects of α-2b interferon on the inflammation and liver fibrosisinduced by DHBV,and to provide evidence for its clinical application.Methods: A total of 40 Beijing ducks (aged 10 d) were randomly divided into control(n=6) and infection(n=34) groups.The ducks in infection group were injected with DHBV positive serum for 3 times in 1 week to establish hepatitis B models.Two weeks later,32 ducks were confirmed infection by using DHBV-DNA test method,and randomly divided into model group (n=10),low dose of α-2b interferon group (n=11, 1×105 U·100 g-1) and high dose of α-2b interferon group (n=11,5×105 U·100 g-1).After 30-day treatment,the serum DHBV-DNA levels of the ducks in various groups were detected by dot hybridization method.The levels of glutamic oxalacetic transaminase (AST),glutamic-pyruvic transaminase (ALT),laminin (LN) and type Ⅲ collagen peptide (PCⅢ) in serum were measured,the levels of IL-6,IL-12,and TNF-α in liver tissue were also measured, and the pathological changes of liver tissue were observed.Results: The level of DHBV-DNA was significantly increased compared with pre-modeling. so the DHBV-infected models were successfully established. Compared with model group,the levels of DHBV-DNA in low and high doses of α-2b interferon groups were significantly decreased (P<0.05);the levels of serum AST and ALT in low and high doses of α-2b interferon groups were significantly decreased (P<0.05); the levels of LN and PCⅢ in high dose of α-2b interferon group were significantly decreased (P<0.05);the levels of IL-6 and TNF-α in liver tissue in low and high doses of interferon groups were decreased (P<0.05); but the IL-12 levels have no significant changes (P>0.05).Compared with model group,the liver fibrosis in low and high deses of α-2b interferon groups was improved.Conclusion: α-2b interferon can relieve the inflammation in duck liver tissue induced by DHBV and liver fibrosis. The study provides the theoretical evidence for further development and clinical application of α-2b interferon.
