Protective effect of celiac transplanting micro-capsulated cells transfected by nerve growing factor gene on 
 hypoxic-ischemic brain damage in new-born |rats

Abstract:

Objective To discuss the protective effect of celiac transplanting micro-capsulated  3T3 cells transfected by nerve growing factor (NGF) gene on  hypoxic-ischemic brain damage(HIBD) in new-born rats,and to provide experimen
tal basis for  HIBD treatment.Methods 60 new-born Wistar rat,were randomly divided into sham operation group (n=20),blank capsule group (n=20),and transplant group (n=20).The rats in sham operation group  were injected with saline.The rat models of HIBD were set up and used in blank capsule group and transplant group.The rats in blank capsule group  were transplanted with blank capsules.The rats in transplant group   were treated by transplanting micro-capsulated  3T3 cells transfected by NGF gene.The changes of water  contents and the apoptotic rates of rat neuron in each group were analyzed.Results The rats in blank caspsule and transplant groups had various  degrees of hydrocephalus,the water content in brain tissues of  rats in blank capsule group was higher than that in sham operation group(P<0.05).The water content in brain tissues of rats in transplant group had no significant difference compared with sham operation  group(P>0.05),but it was lower than that in blank capsule group(P<0.05).The TUNLE immunohistochemistry results showed that there was  significant difference of apoptotic rate of neuron between blank capsule group and sham operation group (P<0.05),and the apoptotic rate of rat neuron in transplant group  was lower  than that in blank capsule group(P<0.05).Conclusion Celiac transplanting of micro-capsulated NGF 3T3 cells  can relieve the hydrocephalus after HIBD and  protect the hippocampal neuron after HIBD.
