Effect of 3-n-butylphthalide on brain-derived neurotrophic factor expression in hippocampal CA1 region of vascular dementia rats

Objective: To study the effect of 3-n-butylphthalide (NBP) on the expression of brain-derived neurotrophic factor (BDNF) in the hippocampal CA1 region of the vascular dementia (VD) rats, and to explore its protective effect on VD. Methods: Eighty healthy Wistar rats were equally and randomly assigned to sham operation group, NBP control group (sham operation + NBP injection), VD group (VD models), NBP treatment group (VD models + NBP injection)(n=20). Each group was divided into four subgroups (n=5):1, 2, 4, and 8 weeks after operation groups. The VD rat models were established by using permanent bilateral common carotid artery ligation. After consciousness, the rats in NBP treatment group and NBP control group were intraperitoneally injected with 5 mg·kg-1·d-1 NBP for consecutive 7 d. The rats in VD and sham operation groups were intraperitoneally injected with 0.2 mL·d-1 saline for consecutive 7 d. At 1, 2, 4, and 8 weeks after operation, the rats in each group were decapitated. The brains were obtained, and then the hippocampus tissues were isolated. The BDNF expression levels in the hippocampal CA1 region were determined using real-time quantitative PCR and immunohistochemistry methods. Results: At 2, 4, and 8 weeks after s operation, the expression levels of BDNF mRNA in the hippocampus of the rats in VD group were significantly higher those in sham operation group(P<0.05);at 4 and 8 weeks after operation, the expression levels of BDNF mRNA in the hippocampus of the rats in NBP treatment groups were significantly higher than that in VD group(P<0.05).The immunohistochemistry results showed that the expression level of BDNF protein in the hippocampal CA1 region of the rats in VD group was higher than that in sham operation group at 4 weeks after operation (P<0.05); the expression level of BDNF protein in the hippocampal CA1 region of the rats in NBP treatment group was higher than that in VD group at 8 weeks after operation (P<0.05). Conclusion: The BDNF expression is increased in the hippocampal CA1 region of the rats with VD after the neurons were injured by ischemia. NBP can increase the BDNF expression level in the hippocampal CA1 region of the VD rats and protect the nerves.

Enhancement of IL-37 in chemosensitivity of cervical cancer HeLa cells to cisplatin

Objective: To transfer the interleukin 37 (IL-37)gene to cervical cancer Hela cells, and to explore the killing effect of IL-37 on the HeLa cells and its enhancement in the chemotherapy sensitivity of HeLa cells. Methods: The pIRES2-EGFP (NC group) and pIRES2-EGFP/IL-37(IL-37 group) plasmids were transfected into the HeLa cells. Q-PCR and Western blotting methods were used to detect the expression levels of IL-37 mRNA and protein. The activities of HeLa cells in NC group, IL-37 group, DDP group and IL-37+DDP group were detected by CCK8 method,and the inhibitory rates of cells were calculated. The gene expressions of signal transducer and activator of transcription 3 (STAT3) and Cyclin D1 were detected by RT-PCR method. Results: Compared with NC group, the expression levels of IL-37 mRNA and protein in IL-37 group were significantly increased(P<0.01). The activities of HeLa cells in DDP(5-15 mg·L-1) groups were inhibited after administration for 24-72 h (P<0.01);the inhibitory rates in IL-37+DDP group were higher than those in DDP group within 48 h after administration (P<0.05).Compared with IL-37 group,the inhibitory rates in IL-37+DDP group was increased with 96 h after administration (P<0.05).Compared with NC group, the expression levels of STAT3 and Cyclin D1 mRNA in IL-37 group were significantly decreased (P<0.01). Conclusion: The over-expression of IL-37 can inhibit the proliferation of cervical cancer cells and enhance the effect of DDP on the chemotherapy of cervical cancer cells which may be related to the down-regulation of the expressions of STAT3 and Cyclin D1 by IL-37.

Regulation effect of endogenous nicotinamide phosphoribosyltransferase on Vimentin expression of glomerular cells in high concentration of glucose

Objective: To investigate the regulation effect of endogenous nicotinamide phosphoribosyl transferase (Nampt) on the Vimentin expression of glomerular cells in high concentration glucose,and to clarify the mechanism of formation of diabetic kidney inflammation fibrosis. Methods: The C57/BL6 diabetic mice were selected and the kidney tissues were collected, and the wild C57/L86 mice were used as control group;the pathological section and tissue fluorescence staining were performed.The expression and location of endogenous Nampt and Vimentin in the glomerular cells were detected by immuno-focused technology. The HBZY-1 cells were randomly divided into 4 groups:low concentration of glucose (LG, 0.56 mmol·L-1) control group,high concentration glucose (HG, 200 mmol·L-1) group,HG + FK866 group and HG+nicotinamide mononucleotide (NMN) group.In HG group, the cells were treated with FK866 (10 μmol·L-1) and NMN (1 mmol·L-1) for 24 h after cultured with HG for 5 d. The expression levels of Nampt, Vimentin, nuclear factor-kappa B p65 (NF-κBp65) and sirtuin type 1 (Sirt1) were detected by immunofluorescence and Western blotting methods. The expression levels of Nampt and Vimentin were detected by RT-PCR and Western blotting methods. Results: The shape and size of glomerulus had obvious atrophy of the mice in severe diabetic group compared with normal C57/BL6 mice group. The expression level of Vimentin in glomerular cells was increased with the increasing of endogenous Nampt (P<0.01). When the HBZY-1 cells were cultured in HG condition,the exprssion levels of Nampt, Vimentin and NF-kB p65 were obviously increased while the Sirt1 expression levels was significantly decreased compared with control group (P<0.01).The expression levels of Nampt, Vimentin and NF-κB p65 in glomerular cells in HG+FK866 HG+NMN groups were singnificartyly decreased compared with control group (P<0.01). Conclusion: The endogenous Nampt over-expression in glomerular cells can enhance the expression of Vimentin under high concentration of glucose stress through NF-κBp65 and Sirt1 signal pathway.

Influence of rat pancreatic islet α cells and its secretions in function of β cells

Objective: To investigate the influence of α cells and glucagon-like peptide 1 (GLP-1) in the function of β cells (INS-1 cells) in the rats, and to elucidate the possible mechanism of α cells and INS-1 cells transplantation in influencing hypoglycemia. Methods: The proliferation abilities of INS-1 cells after treated with 10%, 20% and 30% islet α-cell conditioned medium and 0.03, 0.30, 3.00, 30.0 mg · L-1 of GLP-1 were analyzed by MTT assay. The levels of insulin secretion of INS-1 cells after treated with 10%, 20%, 30% α cells, α-cell conditioned medium and different concentrations of GLP-1 were analyzed by enzyme linked immunosorbent assay (ELISA). The concentrations of Ca2+ in INS-1 cells after treated with high glucose and GLP-1 were analyzed by laser confocal microscope. The expression levels of insulin protein after treated with different concentrations of islet α-cell conditioned medium and different concentrations of GLP-1 were detected by Western blotting methed. After the INS-1 cells, the mixture of INS-1 cells and α cells were transplanted into the left renal capsule of the nude mice, the blood glucose levels and the kidney morphology were observed. The levels of insulin/glucagon in the transplanted cells were detected by immunohistochemistry. Results: Compared with control group,both of α-cell conditioned media and GLP-1 promoted the INS-1 cell proliferation and insulin secretion (P<0.05). The laser confocal microscope results revealed that GLP-1 stimulated the increased intracellular Ca2+ concentration in INS-1 cells (P<0.05). Compared with control group, there was no significant difference in the expression levels of insulin protein in the insulin-1 cells after treated with islet α cell conditioned medium and GLP-1 (P>0.05). Compared with pre-transplantation, the blood glucose level in the transplanted INS-1 cells was significantly decreased at 35 d after renal capsul trasplantation(P<0.05), and even hypoglycemia presented renal capsular in the diabetic nude mice; the transplantation site was obviously swollen. However, the levels of blood glucose had no change of the diabetic rats after transplated with the mixture of INS-1 and α cells(P<0.05).The expression of insulin and glucagon in the INS-1 transplanted cells were found by immunohistochemistry staining. Conclusion: Pancreatic islet α cells and their secretions can promote the INS-1 cell proliferation and insulin secretion, and the mixture of INS-1 cells and α cells transplanted under the renal capsule of the diabetic nude mice can reduce the hypoglycemic effect of INS-1 cell transplantation which might be related to the INS-1 cells that can express both of insulin and glucagon genes.

Effect of N-acetyl cysteine pathway on expression of intermediate conductance Ca2+-activated K+ channels in cardiac fibroblasts of AGT-REN double transgenic hypertension mice

Objective: To investigate the relationship between the level of oxidative stress and the expression of intermediate conductance calcium activated potassium channel (KCa3.1) protein in the cardiac fibroblasts (CFs) during hypertension process,and to clarify the role of KCa3.1 in cardiac fibrosis and its mechanism. Methods: The CFs of male C57B6 and AGT-REN double transgenic hypertension (dTH) mice were cultured and the wild C57B6 mouse CFs were used as control group. The CFs of dTH mice were randomly divided into high blood pressure group (dTH) and N-acetyl cysteine group (NAC). The CFs were treated with different concentrations of NAC for 24 h. The cell proliferation was detected by MTT and double dichlorofluorescein (DCFH-DA) probe was used for the detection of cellular reactive oxygen species (ROS) expression; Western blotting was employed to detect the expressions of collagen Ⅰ, collagen Ⅲ, KCa3.1 channel protein and the changes of PI3K signaling pathway protein phosphorylation. Results: The ROS production and protein expression of KCa3.1 channel of the dTH mice on 4, 8, 12 months were increased compared with 2 months(P<0.05 or P<0.01); the results of MTT suggested that the proliferation rates of CFs were 165.9%,138.72%,110.92% and 109.82% after administration of 1×10-6,1×10-5,1×10-4 and 1×10-3 mol·L-1 NAC in the dTH mice, and 1×10-4 and 1×10-3 mol·L-1 NAC significantly inhibited the proliferation of CFs.Compared with control group,the secretion of collagen Ⅰ and Ⅲ of CFs in the TH mice was decreased in 1×10-4 mol·L-1 NAC group(P<0.01).The results of Western blotting showed that compared with control group, the expression level of KCa3.1 channel protein in CFs of the TH mice in 1×10-4 mol·L-1 NAC group was decreased(P<0.01). Compared with control group,the p-AKT/T-AKt in CFs of the dTH mice was increased (P<0.01); but in NAC group,the p-AKT/T-AKt was lower than that in dTH group (P<0.01). Conclusion: NAC can inhibit the expression of KCa3.1 channel protein in CFs of the dTH hypertensive mice, which may be related to increasing the phosphorylation of AKt/PI3K signaling pathway.

Influence of silencing Smad4 gene in proliferation and apoptosis of breast carcinoma MCF-7 cells

Objective: To study the effects of down-regulated Smad4 expression on the proliferation and apoptosis of breast carcinoma MCF-7 cells, and to explore their mechanisms. Methods: The human breast carcinoma MCF-7 cells and MDA-MB-231cells were cultured in vitro. RT-PCR method was used to detect the expression levels of Smad4 mRNA in MCF-7 cells and MDA-MB-231 cells. The Smad4-shRNA plasmid and Scramble-shRNA plasmid were respectively stably transfected into the MCF-7 cells with high expression of Smad4. The experiment was divided into non-transfected MCF-7 cells (normal control) group,Smad4 gene silencing group, Scramble (negative control)group.The proliferation abilities of the cells in various groups were detected by CCK-8 method. The apoptotic rates of the cells in various groups were detected by flow cytometry. Real-time PCR method was used to detect the mRNA expression levels of the proliferation-related genes CDKN1A, CDK1 and CDK2 and the apoptosis-related genes Suvivin, bcl-2, caspase 3 and caspase 9. Results: The proliferation abilities of cells had no statistical significance between various groups(P>0.05). The mRNA expression levels of CDKN1A, CDK1 and CDK2 in the cells had no statistical significance between various groups (P>0.05).Compared with normal control group and negative control group, the apoptotic rate of the cells in Smad4 gene silencing group was significantly decreased(P<0.01), the expression levels of Suvivin and bcl-2 mRNA in Smad4 gene silencing group were significantly increased(P<0.01), and the mRNA expression levels of caspase 3 and caspase 9 in Smad4 gene silencing group were significantly decreased(P<0.05). Conclusion: Smad4 could induce the apoptosis of MCF-7 cells by down-regulating the expressions of Suvivin and bcl-2 and up-regulating the expressions of caspase 3 and caspase 9.

Improvement effects of Tongfengning Capsule on hyperuricemia and renal function in mice

Objective: To discuss the influence of Tongfengning Capsule(TFN)in the levels of uricacid(UC),creatinine (Cr) and urea nitrogen(BUN) in mouse serum and the activities of the xanthine oxidase(XOD),adenosine deaminase (ADA) activity in the liver homogenate of the mice with hyperuricemia, and to observe the improvement effect of TFN on the pathological changes of liver tissue and to clarify its mechanisms. Methods: The models of mouse hyperuricemia were induced by yeast extract with potassium oxonate. Seventy mice were divided into blank control group,model group, low(200 mg·kg-1), medium(400 mg·kg-1)and high(800 mg·kg-1) doses of TFN groups, allopurinol positive drug control group(50 mg·kg-1),Tongfengshu(TFS,600 mg·kg-1) positive drug control group(n=10). The levels of UC,Cr,BUN in serum and the activities of XOD, ADA in homoggenate were detected and the histopathological changes of the kidney tissue of the mice were measured with HE staining. Results: Compared with blank control group, the levels of serum UC, Cr and BUN of the mice in model group were significantly increased(P<0.01),and the activities of XOD and ADA in liver tissue were also increased(P<0.01). Compared with model group, the levels of serum UC, Cr and BUN of the mice in positive drug control groups and different doses of TFN groups were decreased (P<0.01),and the activities of XOD and ADA in liver tissue were also decreased (P<0.05),especially in high dose of TFN group.Compared with model group,the pathologic changes such as renal glomerulus atrophy, renal interstitial fibrosis and expansion of renal tubule of the mice in positive drug control groups and high dose of TFN group were improved to a certain extent. Conclusion: TFN has improvement effcet on the hyperuricemia in the mice and its mechanism is related to the inhibition of uricogenesis and the promotion of UC excretion.

Anti-fatigue effect of arginylfructosylglucose from red ginseng in mice

Objective: To investigate the anti-fatigue phenomenon induced by forced swimming in the mice,and to explore the anti-fatigue effect of argininyl fructoyl glucose(AFG)from red ginseng in the mice and its mechanism. Methods: The AFG was extracted from red ginseng. The ICR mice were divided into blank control group,low dose of AFG group(100 mg·kg-1),middle dose of AFG group(200 mg·kg-1) and high dose of AFG group(400 mg·kg-1)(n=20). The mice mere given a forced swimming test after continuous gavage for 28 d. The weights,organ indexes,time of forced swimming,contents of lactic acid (LD),blood urea nitrogen (BUN),hepatic glycogen (Gly) and expressing levels of PGC-1α in gastrocnemius of the mice in various groups were detected. Results: Compared with blank control group,the weights and organ indexes of the mice in low,middle and high doses of AFG groups had no significant differences(P>0.05). Compared with blank control group,the time of forced swimming,contents of Gly and expressing levels of PGC-1α of the mice in low,middle and high doses of AFG groups were significantly increased(P<0.05 or P<0.01)in a dose-dependent manner. Compared with blank control group,The contents of LD and BUN in serum of the mice in low,middle and high doses of AFG groups were significantly decreased(P<0.01). Conclusion: AFG has anti-fatigue effect in mice,and its mechanism may be related to energy metabolism.

Establishment of TMJ defect models and evaluation on repair effect of CFR-PEEK material artificial joint in rabbits

Objective: To investigate the method of arthroplasty for reconstructing femporomandibular joint(TMJ) after the establishment of TMJ articular defect models in the rabbits, and to clarify the replacement and repair effects of carbon fiber reinforced polyetheretherketone (CFR-PEEK) material TMJ prostheses, and to provide an experimental basis for the clinical application of CFR-PEEK artificial joints to replace TMJ. Methods: Thirteen healthy adult Japanese rabbits were randomly divided into experimental group (n=6), positive control group (n=4) and negative control group (n=3). The right TMJ articular process of the rabbits in experimental group were resected to establish the joint defect models, and then the CFR-PEEK artificial joints were impanted. The right TMJ articular process of the rabbits in positive control group were resected, and the joint defect models were established without the CFR-PEEK artificial joint implantation. The rabbits in negative control group didn't receive any treatment.The body weights of the rabbits in each group were measured in 13 weeks. The effects of fixation and replacement after artificial joint implantation were assessed by CT imaging. Results: The TMJ articular process defect model was successfully implanted with the CFR-PEEK artificial joint. The CT imaging results after three-dimensional reconstruction and CT images by contrast in vitro showed that the CFR-PEEK-implanted artificial joint was fixed well, it could be instead of the normal joint function.Compared with positive control group, the weights of rabbits in experimental group were significantly increased(P<0.05) at 13 weeks; compared with positive control group, the weights of rabbits in experimental group and negative control group were both significantly increased (P>0.05) at 13 weeks,but there was no significant difference between them(P>0.05).The weights of rabbits in experimental group and negative control group kept normal growth; the weights of rabbits in positive control group was increased slowly, and even stopped growing or was negative. Conclusion: The CFR-PEEK artificial joint can be successfully implanted into the TMJ defect model and be used to repair the defect which help to restore chewing function. The CFR-PEEK is expected to be an ideal material for reconstructing TMJ.

Influence of HPK1 overexpression in proliferation and apoptosis of breast cancer MCF-7 and MDA-MB-231 cells and its mechanism

Objective: To investigate the effects of hematopoietic progenitor kinase 1 (HPK1) overexpression by construction of lentiviral vector on the proliferation and apoptosis of breast cancer MCF-7 and MDA-MB-231 cells, and to elucidate its possible mechanism. Methods: The cells were infected with the lentivirus overxpressing HPK1,and the MCF-7-HPK1 and MDA-MB-231-HPK1 cell lines were stably expressed HPK1;each cell line was divided into three experimental groups:blank group (untreated), control group(empty vector) and HPK1-overexpression group. The expression levels of HPK1 mRNA and protein in breast cancer cells in each group were detected by RT-PCR and Western blotting methods, respectively. The cell proliferation rate was detected by MTT assay. The cell cycle and apoptotic rate were detected by flow cytometry. Transwell assay was used to analyze the cell migration ability. Western blotting method was used to measure the expression levels of caspase 3, PTEN, MMP-9,MMP-2, Ki-67and HPK1 proteins. Results: Compared with blank groups and control groups, the expression levels of HPK1 mRNA and protein in the both cell lines in HPK1 overexpression groups were significantly up-regulated(P<0.05), the proliferation rates were significantly decreased(P<0.05) and the apoptotic rates were significantly increased(P<0.05), the number of cells crossing matrigel was significantly reduced(P<0.05), the cell cycle of MCF-7 was blocked in G1 phase(P<0.05), the expression levels of caspase 3 and PTEN proteins in HPK1 overexpression group were significantly increased(P<0.05), and the expression levels of MMP-2 and MMP-9 proteins were significantly decreased (P<0.05). Conclusion: HPK1 overexpression can inhibit the proliferation and migration of MCF-7 and MDA-MB-231 cells and induce apoptosis, which may be related to the up-regulation of caspase 3 and PTEN and down-regulation of MMP-9, MMP-2 and Ki-67.

Influence of sulforaphane preconditioning in myocardial cold ischemia-reperfusion injury through PI3K/Akt signaling pathway in rats

Objective: To explore the effect of sulforaphane(SFN) preconditioning on the cold myocardial ischemia-reperfusion injury (IRI)in the rats through PI3K/Akt signaling pathway. Methods: Sixty-four health male Sprague-Dawley (SD) rats were randomly divided into cold IRI group, SFN group, LY(LY294002)+cold IRI group, and LY+SFN group(n=16).The allogeneic heterotopic heart transplantation model was established by donor heart into recipient abdomen. The myocardium tissue was taken 24 h after reperfusion for the detection of histological changes using HE staining. The expression levels of Akt, p-Akt, Bax and Bcl-2 proteins were detected by immunohistochemistry and Western boltting methods. Results: The morphological results showed that the myocardium tissue damage was serious in cold IRI group and LY+cold IRI group, it was light in SFN group;the myocardium tissue damage of the rats in SFN+ LY group was ranged between cold IRI group and SFN group. Compared with IRI group, the expression levels of p-Akt protein and Bcl-2 protein in SFN group were increased(P<0.05),and the expression level of Bax protein was decreased(P<0.05). After treatment of blockage LY294002, compared with LY+cold IRI group,the expression level of p-Akt protein in LY+SFN group was not statistically significant(P>0.05), the expression level of Bcl2 protein was increased(P<0.05),the expression levels of Bax protein was decreased), and the ratio of Bcl-2/Bax was also increased(P<0.05). Conclusion: SFN may attenuate cold IRI of heart transplantation through PI3K/Akt signaling pathway in the rats.

Expression of MMP-2 in retina tissue of diabetic retinopathy model rats

Objective: To establish the rat diabetic models by introperitoneal injection of streptozotocin(STZ) to observe the expressions of MMP-2 in retina tissue of the diabetic rats at different periods,and to clarify the effect of MMP-2 in the diabetic retiropathy(DR) of the diabetic rats. Methods: The femal SD rats were divded into normal control group(n=24), 4-week model group(n=30), 6-week model group(n=30) and 8-week model group(n=30).The rats in model groups were intraperitoneally injected with STZ for consecutive 5 d.The rats in normal control group were injected with sodium citrate solution at the same volume. The body weights and blood glucose levels of the rats in each group were measured at 4,6, and 8 weeks.The retina of each rat was removed; RT-PCR and Western blotting methods were used to detect the expression levels of MMP-2 mRNA and protein,and the immunohistochemistry was conducted to observe the morphology. Results: The body weights and blood glucose levels of the rats in each group had no differences(P>0.05) before modeling.Three rats died in 4-week model group, 5 rats died at 6 weeks and 7 rats died at 8 weeks.The body weights of the rats in model group were significantly lower than those in normal control group at the same time(P<0.05 or P<0.01),and the fasting blood glucose levels were signifieantly increased(P<0.05 or P<0.01).The expression levels of MMP-2 mRNA and protein in the retina tissue of the rats in model group were significantly higher than those in normal control group(P<0.05). In normal control group, the retinal structure was clear and the cells were arranged in order, and the ganglion cells were arranged in a single layer; in model group, the retinal tissue structure was loose, the number of ganglion cells were significantly reduced,the inner nuclear layer and rod cell layer cell membrane outside were fuzzy. The MMP-2 positive cells were found to be brown yellow granules, especially in the cytoplasm of ganglion cells and vascular endothelial cells. The positive expression levels of MMP-2 in model group were higher than those in normal control group at 4,6,and 8 weeks. Conclusion: MMP-2 can express in the retina tissue of the diabetic rats.

Inhibitory effect of north-Schisandra chinensis polysaccharide on neutrophil inflammation induced by lipopolysaccharide

Objective: To investigate the effect of Manchu medicine north-Schisandra chinensis polysaccharide (NSCP) on the human neutrophils treated by lipopolysaccharide (LPS) cultivated in vitro, and to elucidate its anti-inflammatory mechanism. Methods: The neutrophilic inflammatory cell model was established with LPS. The experiment included control group, LPS group (1.0 mg·L-1) and NSCP group (1.25, 2.50 and 5.00 g·L-1), the cells in NSCP group were first treated with LPS for 60 min, and then treated with different concentrations of NSCP. The levels of TNF-α in neutrophils were measured with ELISA and the apoptotic rates were detected by flow cytometry. Results: The level of TNF-α in LPS group was increased compared with control group (P<0.05). The level of TNF-α in NSCP group was decreased compared with LPS group (P<0.05).The apoptotic rate in LPS group was decreased compared with control group (P<0.05);the apoptotic rates in NSCP group were increased with the increasing of time and dose, and the best effect was found 16 h after treatment with 5 g·L-1NSCP;the apoptotic rate in NSCP group was significantly increased compared with LPS group (P<0.05). Conclusion: NSCP can perform the anti-inflammation effect through the suppression of LPS-induced TNF-α secretion in neutrophils and the promotion of neutrophils apoptosis.

Influence of astaxanthin in refractive status and pathohistology of myopia models caused by concave lens of guinea pigs

Objective: To investigate the effect of astaxanthin on the refractive status and pathological morphology of the guinea pig myopia models induced by concave lens, and to elucidate the improvement effect of astaxanthin on the refractive status of myopia. Methods: A total of 48 healthy guinea pigs were randomly divided into blank control group(n=12) and model group(n=36);the guinea pig myopia models(right eyes) were established with far vision optical method(left eyes were used as control).The guinea pig models were divided into model control group, high of astaxanthin (50 ig·kg-1) group and low dose of astaxanthin(25 ig·kg-1)group. After 2 and 4 weeks treatment, the diopters and the lengths of optic axis of both eyes of each guinea pig in various groups were tested, and at 4 weeks the right eyes of the pigs were removed and the pathomorphology was observed by HE staining. Results: The diopters of the model eyes in model group were lower than those of left eyes used as control(P<0.05) and those of right eyes in blank control group after 2 and 4 weeks (P<0.05 or P<0.01).The length of optic axis of the model eye in model group was increased compared with left eye used as controls and blank control group after 2 weeks (P<0.05) and especially at 4 weeks(P<0.01). Compared with model group,the axial oculi and diopters of the guinea pigs in astaxanthin groups were increased (P<0.05);the pathohistological changes were improved,the collagen fracture was obviously decreased,and the collagen fibers were orderly arranged. Conclusion: Astaxanthin can recover the length of optic axis, diopter and histomorphology of sclera of guinea pig myopia models in some extents.

Influence of autophagy inhibited by chloroquine in apoptosisof myelogenous leukemia cells promoted by decitabine

Objective: To study the influence of chloroquine combined with decitabine in the apoptosis of leukemia K562 cells and KG-1a1Aor1a cells, to explore the effect of autophagy on the leukemia cell apoptosis induced by decitabine, and to clarify its mechanism. Methods: The leukemia K562 and KG-1a1Aor1a cells were cultivated in vitro and divided into blank control group, decitabine group (10 μmol·L-1) and chloroquine(50 μmol·L-1) combined with decitabine group (combined group). The leukemia cells in combined group were pre-treated with chloroquine for 6 h before experiment. After treatment with drugs for 24 and 48 h, the number of cells was detected and by CCK-8 method the inhibitory rates of proliferation cells were calculated;the apoptotic rates and mitochondrial membrane potential were detected by flow cytometry. Q-PCR method was carried out to determine the gene expression levels of Atg7 and Atg12, and Western blotting was used to test the protein expression of LC3. Results: After treatment for 24 and 48 h, the number of K562 and KG-1a1Aor1a cells in decitabine group and combined group were decreased compared with blank control group(P<0.05 or P<0.01); the apoptotic rates and mitochondrial membrane potential were remarkably increased (P<0.05 or P<0.01). Compared with decitabine group, the number of K562 and KG-1a1Aor1a in combined group was significantly decreased, and the apoptotic rates were remarkably increased (P<0.05). After treatment for 24 h, the expression levels of Atg7, Atg12 and LC3-Ⅱ/LC3-Ⅰ in the leukemia K562 and KG-1a1Aor1a cells in decitabine group were significantly increased compared with blank control group (P<0.05 or P<0.01);the expression levels of Atg7, Atg12 and LC3-Ⅱ/LC3-Ⅰ in the leukemia K562 and KG-1a1Aor1a cells in combined group were significantly decreased compared with decitabine group (P<0.05 or P<0.01). Conclusion: Decitabine could promote the apoptosis of leukemia cells, and the inhibition of autophagy by chloroquine can promote the apoptosis induced by decitabine.

Objective: To investigate the therapeutic effect of folic acid combined with vitamin B12 on atherosclerosis induced by hyperhomocysteinemia in the rats, and to elucidate its therapeutic mechanism. Methods: Thirty-three rats were randomly divided into control group, model group and treatment group (n=11).The rats in control group were given normal diet,the rats in model group were given normal diet and methionine,and the rats in treatment group were given normal diet, methionine, perfused folic acid and vitamin B12. The histopathological changes of thoracic aorta of the rats were observed by HE staining. The levels of serum Hcy and matrix metalloproteinase-9 (MMP-9) were detected with ELISA,and the expression levels of CCR5 and CC chemokine ligand 5 (CCL5) proteins in thoracic aortic smooth muscle cells were detected by immunohistochemical staining. The expression levels of CCR5 and CCL5 mRNA in thoracic aortic smooth muscle cells were detected by RT-PCR. Results: The results of HE staining showed that the medial smooth muscle cells of the rat thoracic aorta were arranged neatly in control group, the intimal elastic membrane was complete and the endothelial cells were continuous; the smooth muscle cells of the thoracic aorta were loose in model group, the endometrium was damaged, the foam cells were formed under the intima, and the endothelial cells were necrotic;the pathological changes of the thoracic aorta of the rats in treatment group were between control group and model group. The levels of serum Hcy of the rats in model group and treatment group were higher than that in control group (P<0.05); the serum Hcy level in treatment group was lower than that in model group (P<0.05).The levels of serum MMP-9 of the rats in model group and treatment group were higher than that in control group (P<0.05);the serum MMP-9 level in treatment group was lower than that in model group (P<0.05).The expression gray values of CCR5 and CCL5 proteins in the thoracic aortic smooth muscle cells of the rats in rats in model group and treatment group were lower than those in control group (P<0.05); the expression gray values of CCR5 and CCL5 proteins in the thoracic aortic smooth muscle cells of the rats in treatment group were higher than those in model group (P<0.05).The relative expression levels of CCR5 and CCL5 mRNA in the thoracic aortic smooth muscle cells of the rats in model group and treatment group were higher than those in control group (P<0.05);the relative expression levels of CCR5 and CCL5 mRNA in the thoracic aortic smooth muscle cells of the rats in treatment group was lower than that in model group (P<0.05). Conclusion: Folic acid combined with vitamin B12 could decrease the serum Hcy and MMP-9 levels and the expression levels of CCR5 and CCL5 in the thoracic aortic smooth muscle cells which may be one of the mechanisms of folic acid combined with vitamin B12 in the treatment of atherosclerosis.

Construction and eukaryotic expression of recombinant HSA-TP5 fusion gene expression vector

Objective: To construct the expression vector of the fusion protein of human serum albumin (HSA) and thymopentin (TP5) and to express it in Pichia pastoris, and to elucidate the biological activity of fusion protein. Methods: The HSA-TP5 fusion gene was constructed by gene recombination and transfected into Pichia pastoris to construct the eukaryotic expression system of HSA-TP5. The recombinant eukaryotic expression plasmid of PPICZα -HSA-TP5 was obtained by agarose gel electrophoresis and purification reagent. The two step fermentation method was used to ferment gene engineering bacteria of HSA-TP5 in high density, and the fermentation supernatant protein was precipitated and concentrated; the purified fusion protein was obtained by cation exchange chromatography and hydrophobic chromatography and analyzed by SDS-polyacrylamide gel electrophoresis. The effect of the fusion protein on the proliferation of lymphocytes was detected by MTT assay. Results: The HSA target gene fragment with length of 1 845 bp was achieved by PCR method. The HSA-TP5-pPICZαC fusion plasmid was identified by restriction endonuclease digestion, and the fragment length was 707 bp. The sequence analysis showed that the HSA and TP5 sequences of the target genes were identical with the gene sequences reported in GenBank and were fused by forward fusion. PCR method confirmed that the eukaryotic recombinant plasmid PPICZ αC -HSA-TP5 was integrated into the yeast genome, and compared with control group, the target gene PCR product length was found to be 1 860 bp. SDS-PAGE analysis showed that the expression level of HSA-TP5 fusion protein was gradually increased with the induction time within 72 h. HSA-TP5 fusion protein was purified by cation exchange chromatography and AKTA multifunctional protein purification system.The MTT assay results showed that HSA-TP5 fusion protein was consistent with TP5 protein in promoting lymphocyte proliferation activity. Conclusion: HSA-TP5 fusion protein can be obtained by constructing the eukaryotic expression system of Pichia pastoris and owns the biological activity.

Effect of icariine combined with embryonic stem cells on apoptosis of kidney epithelial cells in contrast-induced nephropathy rat models

Objective: To investigate the effect of icariin combined with embryonic stem cells on the renal epithelial cells in the contrast-induced nephropathy(CIN) rats and its possible mechanism, and to provide experimental evidence for clinical application of icariin. Methods: Fifty SD rats were randomly divided into,control group, model group, icariin group, stem cell group and combination group(n=10).The rat models were established by tail vein injection of INDO, L-NAME and diatrizoate. The serum creatinine (Scr) levels of the rats were detected before and after modeling.The expressions of p38MAPK and caspase-3 mRNA and protein were detected by semi-quantitative RT-PCR and Western blotting method. The apoptotic rate of renal epithelial cells was detected by flow cytometry. Results: The levels of Scr of rats were significantly increased 48 h after combination contrast agent injection, suggesting that the CIN models were successfully set up.Compared with model group,the Scr level of the rats in group was significantly decreased (P<0.05). compared with model group the expression levels of p38MAPK and caspase-3 mRNA and protein in icariin group and stem cell group and in combination group were decreased (P<0.05).The apoptotic rate of renal epithelial cells in combination group was significantly lower than those in icariine and stem cell groups (P<0.05). Conclusion: Icariine combined with embryonic stem cells has a protectice effect on the renal epithelial cells in the CIN rats, and its mechanism may be related to the inhibition of the related factors in the apoptosis pathways.

Clinical significance of IL-17 expression in serum of patients with psoriasis vulgaris and effects of shikonin on IL-6 and IL-23 production in IL-17-stimulated HaCaT cells

Objective: To explore the expression level of serum interleukin-17 (IL-17) in the patients with psoriasis vulgaris and the changes of interleukin-6 (IL-6) and interleukin-23 (IL-23) levels secrered by the HaCaT cells after stimulated with IL-17, and to clarify its clinical significance and the intervention effect of shikonin. Methods: Twenty-five normal controls, 29 patients with psoriasis vulgaris and different groups of HaCaT cells (blank control group,IL-17-24 h group,IL-17-36 h group,IL-17-48 h group,shikonin+IL-17 group,CsA+IL-17 group and IL-17 group) were used as the subjects. The levels of serum IL-17 in the patients with psoriasis vulgaris and the levels of IL-6 and IL-23 in supernatant of HaCaT cells in various groups were measured by double antibody sandwich enzyme-linked immunosorbent assay(ELISA) and real-time polymerase chain reaction RT-PCR was used to detect the expression levels of IL-6 and IL-23 p19 mRNA in HaCaT cells in various groups.At the same time,Cell Counting Kit-8 (CCK-8) method was used to detect the viabilities of HaCaT cells in various groups. Results: The level of serum IL-17 of the patients in psoriasis vulgaris group was increased compared with nomral control group, especially in the severe skin lesion group(P<0.05). The levels of IL-6 and IL-23 in HaCaT cells and supernatant and their mRNA expression levels in IL-17-24 h,IL-17-36 h and IL-17-48 h groups were significantly higher than those in blank control group (P<0.01). The expression levels of IL-6 and IL-23 in HaCaT cells and supernatant and their mRNA expression levels in shikonin+IL-17 and CsA+IL-17 groups were lower than those in IL-17 groups (P<0.05). No statistical differences were found in the cell viabilities between each drug treatment group and blank control group (P>0.05). Conclusion: The expression level of IL-17 in the patients with psoriasis vulgaris is significantly increased,especially in the patients with severe psoriasis vulgaris.IL-17 can promote the secretion of IL-6 and IL-23 in HaCaT cells in a time-dependent manner. Shikonin can inhibit the proinflammatory effect of IL-17.

Observation on short-term efficacy of thermal-therapy combined with 125I particles in treatment of recurrent rectal cancer

Objective: To evaluate the clinical efficacy of thermal-therapy combined with 125I radioactive particles by comparing the results of preoperative and postoperative images,serum carcino-embryonic antigen(CEA) and following-up after treatment of the patients with recurrent rectal cancer. Methods: Nineteen rectal cancer patients with postoperative recurrence after radiotherapy were treated with 125I implantation guided by CT (20 cases were treated with operation, but 1 patient was exited who couldn't tolerate the thermal-therapy). Radioactive particle treatment planning system (TPS) was used to make the plan before the particle implantation, and the distance of the particle was 1.0 cm; all patients received CT scanning and were quantificationally evaluatated after implantation; the number of seeded particles was about 12-58, the radioactivity of the 125I particle was 0.5 mCi, and the matching dose surrounding the tumor was 90-140 Gy. All patients were treated with microwave thermal-therapy after particle implantation, 60 min per time, lasting the temprature at 43℃, twice per week, for three weeks. And the patients were followed up after treatment for 3 months, the efficacy was evaluated by image and CEA results, and the urinary frequency, dysuria, hematuria and rectal bleeding were evaluated. Results: Compred with before treatment, after following up for 6 months, the image results showed that the tumor volume was reduced, and the blood CEA level of all patients were decreased from (30.25±8.32)mg·L-1 to (11.89±5.22) mg·L-1 (t=13.158,P<0.01);the local efficacy was 94.7% (18/19),and the pain relief rate was 94.4% (17/18);the NRS median was 6(4,7)before treatment,and it was 1(0,3) after treatment, there was significant difference (P<0.001).There were no frequent urination, urinary pain, hematuria, rectal bleeding and other complications in all patients. Conclusion: Thermal-therapy combined with 125I radioactive particle implantation has good curative effects on the recurrent rectum cancer, and is effective means for recurrent rectum cancer treatment.

Meta-analysis on correlation between oral lichen planus and Candida albicans infection

Objective: To evaluate the association of oral lichen planus(OLP) with Candida albicans infection by Meta-analysis, and to provide basis for the antifungal treatment of OLP. Methods: Bibliographic searches were conducted in PubMed, EMBase, Medline, The Cochrane Library, CNKI,VIP, and Wanfang databases from inception to December 30 in 2016. The literatures were screened according to the inclusion and exclusion criteria by two reviewers independently, the methodology quality was evaluated after data abstraction, and then the RevMan 5.3 and Stata 12.0 software were used for Meta-analysis to calculate the crude odd ration (OR) and 95% confidence intervals (95% CI). Funnel plot and Egger's methods were used for the evaluation on the publication bias of papers. Results: Thirteen trials were included in this study. Collectively, 1 073 OLP patients and 766 controls were available for analysis. The detection rate of Candida albicans in the OLP patients was significantly higher than that in healthy controls (OR:2.30, 95% CI:1.60-3.31, P<0.01). Seven studies of these included literatures were performed on clinical typing of the OLP patients, and the risk of Candida albicans infection in the patients with erosive-atrophic OLP (E-OLP) was significantly increased(OR=10.12,95%CI:4.05-25.28,P<0.01). Conclusion: OLP has a clear association with Candida albicans infection, and this association is particularly significant in the E-OLP patients.

Clinical characteristics and drug resistance analysis of non-fermentative bacteria infection in infants

Objective: To study the clinical characteristics, prognosis and drug resistance caused by the non-fermenting bacteria in the infants, and to provide reference for the doctors to recognize the infection features and its treatment. Methods: A total of 91 cases of non-fermentative bacteria infection were selected and the clinical materials were retrospectively analyzed. The clinical data and prognosis of the pediatric patients were analyzed, as well as the distribution and drug resistance of non-fermentative bacteria.The bacterial resistance genes were detected by PCR method, and the positive results were analyzed by gene sequencing. Results: In the past 5 years, the non-fermentative bacteria strains were isolated in 91 infant patients, including 35 cases of newborn (19 cases were premature infants), 29 cases aged less than 1 year old, 27 cases aged from 0 year to 3 years old. Among these patients, 60 were male and 31 were female. There were 41 cases with underlying diseases (45.05%), 16 cases with organ dysfunction(17.58%), 3 cases discharged automatically(3.29%),and 1 case dead(1.09%). A total of 102 strains of non-fermentative bacteria included 42 strains of Pseudomonas aeruginosa, 33 strains of Acinetobacter baumannii, 21 strains of Stenotrophomonas maltophilia and 6 strains of other non-fermentative bacteria. Forty-four strains were isolated from neonatal ward, 33 strains (32.35%) from neonatal ICU (43.13%), 25 strains (24.50%) were isolated from general pediatric ward. These strains were mainly from respiratory tract secretions and blood samples, nearly 84.31%. The isolation rates of MDR, XDR, PDR Acinetobacter baumannii and Pseudomonas aeruginosa were 63.63% and 19.04%, respectively. There were 40.48% of Pseudomonas aeruginosa isolates were resistant to imipenem, blaPER had the highest positive gene rate (28.57%).There were 36.36% of Acinetobacter baumannii isolates were resistant to imipenem, all resistant strains carried blaOXA-51 and blaOXA-23 genes. Conclusion: The infants with underlying diseases or invasive diagnosis and treatment are easy to infect non fermentative bacteria. Most strains of them are drug-resistant and difficult to be treated with long duration and high risk.

Expression of O-GlcNAc transferase in hepatocellular carcinoma tissue and its clinical significance

Objective: To investigate the expressions of O-GlcNAc transferase (OGT) in the hepatocellular carcinoma (HCC) tissue, cells and serum, and to clarify the significance of OGT in diagnosis and treatment of HCC. Methods: Western blotting method was used to detect the expression levels of OGT in 20 samples of HCC tissue and matched non-tumor tissue, 8 hepatoma cell lines and normal liver (human) cell lysate. The expression levels of serum OGT were detected by direct ELISA in 20 liver cirrhosis (LC) patients, 20 HCC patients and 20 healthy volunteers.The relationship between the expression levels of protein in HCC tissue,cells and serum and the occurrence and development of tumor were analyzed. Results: The Western blotting results showed there were expressions of OGT in both HCC tissue and matched non-tumor tissue; The positive expression rate of OGT in HCC tissue was significantly higher than that in non-tumor tissue (P<0.05).There were expressions of OGT in both hepatoma cell lines and normal liver (human) cell lysate; the positive expression rate of OGT in HCC cell lysate was significantly higher than that in the normal liver cell lysate(P<0.05).The result of direct ELISA showed that the serum OGT levels of the LC and HCC patients were equally higher than that of the healthy volunteers (P<0.05). Conclusion: OGT highly expresses in HCC tissue and OGT is a promising biomarker for the diagnosis of HCC.

Objective: To measure the morphologic data of Chinese temporomandibular joint (TMJ) with computed tomography (CT) reconstruction technology, and to clarify the safe area for the fixation of Chinese temporomandibular joint prosthesis. Methods: A total of 100 adult skulls were scanned by CT and reconstructed.The width,thickness and angle h1,h2,h3,h4,d1,d2,d3,d4,ab,bc,cd,ad,αandβof zygomatic arch,the width and height BD and h of articular fossa and the width,thickness and angle S,h5,h6,h7,l1,l2,l3,l4,l5,m1,m2,m3,m4,m5 and γ of mandible in the fixed area of the TMJ prosthesis were measured; all the measurement indexes were divided into left and right groups by direction, and SPSS 19.0 software was used for the comparion of the measurement between left and right groups. Results: There were no statistical differences in the measurement index values of h1,h2,h3,h4,d1,d2,d3,d4,ab,bc,cd,ad,α,β,BD,h,S,h5,h6,h7,l1,l2,l3,l4,l5,m1,m2,m3,m4,m5 and γ in zygomatic arch, articular fossa and mandible in the fixed area of the TMJ prosthesis between left and right groups (P>0.05); the width of h2 was longer than that of h3 in zygomatic arch (P=0.048); the thickness of d3 was thicker than that of d4 (P<0.01). Conclusion: The measurement data of TMJ fixed area is obtained with three-dimensional CT images.

Improvement effect of aerobic exercise on haemodynamics and exercise endurance of male old patients with simple obesity

Objective: To investigate the effects of 12-week low intensity aerobic exercise on the body composition, haemodynamics and exercise endurance of the male old patients with simple obesity, and to provide evidence for conducting special exercise prescription. Methods: Forty-six male old simple obesity patients were divided into control group (n=19) and exercise group (n=27) according to subjective will. The subjects in control group maintained daily life while those in exercise group performed a 12-week low intensity aerobic exercise. Before and after experiment, the rest heart rates and blood pressures were measured, the maximal oxygen intake (VO2max), maximal power (MP) and exhaust time (ET) were detected by symptom limited graded exercise test, the heart rate curve and blood pressure curve were depicted, and the body composition was determined by body composition analyzer. Results: Compared with before experiment, the fat mass, the percentage of fat body, rest heart rate and blood pressure of the patients in exercise group were decreased (P<0.05), the VO2max, MP and ET were increased (P<0.05), and the heart rate curve and blood pressure curve during exercise test shifted right; but the indicators in control group showed no significant changes (P>0.05). Compared with control group, the fat mass, percentage of body fat, rest heart rate and blood pressure were decreased (P<0.05) and the VO2max, MP and ET were increased (P<0.05) in exercise group after experiment. Conclusion: Low intensity aerobic exercise of 12-week may effectively improve the haemodynamic abnormality, reduce the body composition and enhance the exercise endurance.

Accuracy verification and reliability analysis of three-dimensional printing model in orthopedics and maxillofacial surgery

Objective: To discuss the accuracy of stereolithography (SLA) -3D printing model in orthopedics and maxillofacial surgery, and to clarify the reliability of its clinical application. Methods: A total of 33 cases of SLA-3D printing models were randomly included(10 long bones, 13 irregular bones and 10 prostheses), and the CT scanning and three-dimensional reconstruction were performed. According to the clinical application characteristics of the models, the max height (MH), max length (ML), max width (MW), width of the trochlear (WT), length of the trochlear (LT), length of the glenoid cavity (LGC), width of the glenoid cavity (WGC), distance of pubic tubercle-anterior superior iliac spine (DPI), diameter of the acetabulum (DA), diameter of the marrow cavity (DMC), anterior height of right ramus (AHRR) and posterior height of right ramus (PHRR) were selected to measure the original data and model data, respectively. The absolute/relative errors were calculated. Intraclass correlation coefficient (ICC) was used to evaluate the consistency of the data.The three-dimensional error analysis of the prosthesis model was carried out. Results: There were no significant differences in the anatomical parameters except MW of irregular bone (P>0.05) in 33 cases of SLA-3D printing model.The values of ICC were all greater than 0.950; the maximum values of absolute/relative errors were -0.58 mm and -1.37%, respectively; the maximum 3D error of prosthesis model was 0.237 mm, and the average value was 0.132 mm. Conclusion: The CT data of SLA-3D model is highly consistent with the original data, and it is accurate and reliable in the diagnosis and treatment of the diseases in orthopedics and maxillofacial surgery.

Influence of docetaxel combined with cisplatinand single-agent cisplatin concurrent chemoradiotherapy in prognosis of patients with advanced cervical cancer and evaluation on security

Objective: To investigate the influencing factors of prognosis in the patients with advanced cervical cancer, and to evaluate the influence of single-agent cisplatin and docetaxel combined with cisplatin concurrent chemoradiotherapy in the prognosis of the patients and the security, and to provide the basis for clinical treatment of advanced cervical cancer. Methods: A total of 218 patients with advanced cervical cancer who received the single-agent cisplatin or docetaxel combined with cisplatin concurrent chemoradiotherapy were enrolled as the subjects. The patients were divided into two groups according to their chemoradiotherapy treatments:single-agent group and docetaxel combined with cisplatin concurrent group.All the patients had received external beam radiation therapy combined with high-dose rate intracavitary brachytherapy.A follow-up in 218 patients was performed until January 2017.The information about clinical characteristics including age, tumor diameter, clinical stage, chemotherapy treatments, pathological grade, pathological type, primiparous age and adverse effects after treatment were collected by telephone follow-up,outpatient service and reexamination.The differences in the clinical characteristics of the patients were compared between two groups,the association between the clinicopathological characteristics and the prognosis of the patients was analyzed, and the security of these two treatments was evaluated. Results: There were no statistical differences in the age, tumor diameter, clinical stage, pathological grade, pathological type and primiparous age of the patients between two groups(P>0.05).The 1-year, 3-year and 5-year overall survival (OS) rates of 218 patients were 96.33%, 76.15% and 63.76%, respectively.The 5-year OS rate of the patients in docetaxel combined with cisplatin group was higher than that of the patients in single-agent cisplatin group (χ2=8.032,P=0.005).The univariate analysis results indicated that tumor diameter, clinical stage, chemotherapy treatments, and adenocarcinoma were the potential predictive factors(P<0.05). The Cox regression analysis results showed that clinical stage[stage Ⅲ:P=0.016, HR(95%CI)=1.90(1.13-3.19)[stage Ⅳ:P<0.001, HR(95%CI)=19.13(7.84-46.68)], chemotherapy treatments[P=0.009, HR(95%CI)=0.54(0.34-0.86)] and adenocarcinoma pathological type[P=0.021, HR(95%CI)=1.88(1.10-3.21)]were the prognostic risk factors of the patients,the patients in clinical stage ⅡB,received docetaxel combined with cisplatin concurrent chemoradiotherapy and with pathological types of squamous cell carcinomas had a better outcome. Compared with docetaxel combined with cisplatin group, leucopenia (Z=-2.060,P=0.039) and neutropenia (Z=-2.246, P=0.025) in single-agent cisplatin group were lighter. Conclusion: Clinical stage,chemotherapy treatments and adenocarcinoma were the independently prognostic factors of the patients with advanced cervical cancer.The patients receive docetaxel combined with cisplatin concurrent chemoradiotherapy have a better prognosis and no severer adverse effects compared with single-agent cisplatin treatment.

Objective: To explore the positive expressions of biological markers human mammaglobin(hMAM)combined with matrix metallopeptidase 9(MMP-9) and human epidermal growth factor receptor 2(C-erbB2)mRNA in peripheral blood of the breast cancer patients with micrometastases,and to clarify its clinical application value in diagnosis of the micrometastases in peripheral blood of the breast cancer patients. Methods: A total of 74 patients with breast cancer,21 patients with breast fibroadenoma and 10 healthy controls were selected as the subjects. All the patients received surgical treatment and the peripheral blood was collected.The mRNA expression levels of hMAM,MMP-9 and C-erbB2 in peripheral blood were measured by the real-time fluorescent quantitative PCR.The positive expression rates of detection of hMAM,MMP-9 and C-erbB2 were compared,and the differences in detection of hMAM combined with MMP-9 and C-erbB2 between the patients with different clinicopathologic features were analyzed. Results: In the breast cancer patients with lymph node metastasis,the differences of positive expression rates ofMMP-9 and C-erbB2 mRNA were significant(χ2=6.450, P<0.05;χ2=5.636, P<0.05),and the difference of positive expression rate of hMAM mRNA was sigificant between HER-2 positive and negative patients(χ2=5.804, P<0.05).The positive expression rates ofindividualhMAM and combined with MMP-9 and C-erbB2 were 37.84%(28/74),59.46%(44/74)and 48.65%(36/74)in the breast cancer patients,the combined postive expression rate of these three kinds of markers was 64.86%(48/74),which were higher than those in healthy controls group(χ2=5.676,P<0.05;χ2=3.102,P>0.05;χ2=5.339,P<0.05;χ2=2.310,P>0.05),fibroadenoma ofbreast group(χ2=8.438, P<0.01; χ2=4.491, P<0.05; χ2=7.982, P<0.01; χ2=4.844, P<0.05)and non-breast cancer group(healthy controls group+ breast fibroadenoma group)(χ2=13.093, P<0.01; χ2=6.471, P<0.05; χ2=11.837, P<0.01;χ2=6.103, P<0.05).The positive expression rates ofindividualhMAM and the joint detection in the breast cancer patients at stage Ⅲ+Ⅳ were higher than those in the patients at stageⅠ+Ⅱ;the positive expression rates of individualhMAM and combined with C-erbB2 were statistically significant(χ2=5.157, P<0.05; χ2=4.912, P<0.05). Conclusion: hMAM has a low positive rate in the diagnosis of micrometastases in the breast cancer patients,while hMAM combined with MMP-9 and C-erbB2 detectioncould improve the positive rates.which presents some clinical application value for the early diagnosis of breast cancer micrometastases.

A case report of breast cancer complicated with thyroid cancer and dermatomyositis and literature review about relationships between three kinds of diseases

Objective: To provide clinicians with insights about the patients with breast cancer complicated with thyroid cancer and dermatomyositis, and to improve early detection of the diseases by observing the clinical characteristics of 1 case of breast cancer complicated with thyroid cancer and dermatomyositis and reviewing the literatures about the relationships between these three kinds of diseases. Methods: The medical information of this patient, including gender, age, clinical manifestations, glucocorticoid treatment dose, type of concurrent tumor and the time point it occured and therapeutic regimen were collected and recorded. These clinical data were retrospectively analyzed. Results: The patient with dermatomyositis was diagnosed with breast cancer and thyroid cancer in succession. Oral administration of 50 mg dexamethasone per day was continued in the treatment of dermatomyositis. Then the patient received 4 cycles of pirarubicin/cyclophosphamide(AC) followed by 4 cycles of paclitaxel/Hessaitin (TH) as neoadjuvant chemotherapy for breast cancer. During the 24 weeks of chemotherapy,the breast tumor size was gradually decreased while there was no significant change in thyroid tumor size. The clinical symptoms of dermatomyositis were also improved. The blood lactic acid dehydrogenase and alpha hydroxybutyrate dehydrogenase levels were decreased, but not obviously. After 8 courses of AC-TH neoadjuvant chemotherapy followed by radical resection of thyroid cancer, there was no significant improvement in the symptoms of dermatomyositis 1 week after operation and the myocardial enzyme levels remained unchanged. Then modified radical mastectomy was performed. The myocardial enzymes were examined again 1 week after the second operation all of them were decreased to the normal levels.The clinical symptoms of dermatomyositis were also improved. Conclusion: Although the relationships between the three diseases is still controversial, the clinical data of the patient and relevant literatures collected in this paper support that breast cancer is associated with thyroid cancer and dermatomyositis is associated with breast cancer, but not thyroid cancer.

Lung adenocarcinoma treated by Erlotinib HCL Tablets and chemotherapy combined with bevacizumab after drug resistance:A case report and literature review

Objective: To report a case of lung adenocarcinoma of stage Ⅳ with epidermal growth factor receptor(EGFR) mutation treated by Erlotinib HCL Tablets and chemotherapy combined with bevacizumab after drug resistance, and to review the related literatures. Methods: A female patient without smoking history was diagnosed with lung adenocarcinoma (stagecT3N2M1 Ⅳ) with metastatic lesions in liver and bones by chest CT, lung puncture biopsy,and whole-body PET-CT. EGFR gene test reported mutation (19 del). Through treatment of overall survival, pemetrexed disodium for injection plus carboplatin injection, gemcitabine hydrochloride for injection plus cisplatin injection, paclitaxel for injection(albumin bound) single and with bevacizumab; the overall survival(OS) was assessed as 32 months. And the period of Erlotinib HCL Tablets showed 10 months of progressive-free survival(PFS). However, due to the metastatic lesions that suppressed biliary system in liver, hyperbilirubinemia emerged(total bilirubin 304 μmol·L-1, direct bilirubin 193 μmol·L-1). Bevacizumab was adopted and the syndrome relieved(total bilirubin 70 μmol·L-1, direct bilirubin 35 μmol·L-1), yielding three months of PFS. But the bilirubin level upgraded with total bilirubin of 908 μmol·L-1. The patient died because of hyperbilirubinemia. Results: 10-month PFS occupied 1/3 of OS was acquired in the patients with EGFR-mutated lung adenocarcinoma(stage Ⅳ) after treated with Erlotinib HCL Tablets. Chemotherapy combined with bevacizumab acquired 3 months of PFS after hyperbilirubinemia emerged with drug resistance and multi-lines of chemotherapy. Conclusion: EGFR-TKIs are effective in treatment of lung adenocarcinoma with EGFR mutation; bevacizumab can inhibit the formation of vessels by targeting on VEGF under the circumstance of multiple drug resistance. Therefore, chemotherapy combined with bevacizumab can be used as the fifth-line of therapy in dealing with the hyperbilirubinemia induced by lung adenocarcinoma with liver metastasis.

Clinical analysis on sudden sensorineural hearing loss complicated with BPPV and literature review

Objective: To summarize the clinical characteristics of the patients suffering from sudden senserineural hearing loss (SSNHL) with benign paroxysmal positional vertigo (BPPV), and to investigate the possible pathogenesis. Methods: The clinical materials of thirsty-five patients with SSNHL complicated with vertigo were retrospectively analyzed.The clinical information, including the traits of vertigo and prognosis of hearing loss of the patients in SSNHL with BPPV group (n=9), and SSNHL with non-BPPV group (n=26) were compared. Results: All patients had profound hearing loss and unsatisfied prognosis of hearing loss, and the response rate was 17.1%,the effective rate was 11.4%,and the curative rate was 5.7%. 25.7% of all cases were diagnosed of BPPV by medical history and postural tests, who had heavier hearing loss than the others. And BPPV comlicated with SSNHL was often found within 1 week after the hearing loss (average value:4.1 d), and all on the same side with the hearing loss ear. BPPV tended to happen in total deafness(6 cases,66.7%)and flat type(3 cases,33.3%),and were often involved in horizontal semicircular canal (6 cases,66.7%) and posterior semicircular canal(3 cases,33.3%). The patients with SSNHL complicated with BPPV needed more times of reduction maneuver (average value:2.3 times), with a satisfied therapeutic effect for reducing the vertigo of the patients. Conclusion: SSNHL complicated with BPPV has a high morbidity in the patients with heavy hearing loss, and often affect the horizontal semicircular canal, which needs the repeated reduction maneuver.

AIDS complicated with adrenal insufficiency:A case report and literature review

Objective: To explore the clinical characteristics,diagnosis,and treatments of adrenal insufficiency (AI) in the patients with acquired immune deficiency syndrome (AIDS),and to improve the clinician's understanding of the disease and provide evidences for its diagnosis and treatments. Methods: The clinical data of one patient with AIDS and AI were retrospectively analyzed and the diagnosis and treatments were summarized,and the relative literatures were reviewed. Results: The patient was clearly diagnosed as AIDS and AI after relevant examinations.The symptoms such as fatigue, nausea and vomiting of the patient were disappeared,the food intake of the patient was increased,and the electrolyte was normal after some treatments such as hormone replacement therapy(oral prednisone acetate 5.0 mg at 8:00 am. and 2.5 mg at 4:00 pm.) and symptomatic treatments.The patient died of a severe opportunistic infection when followed up for 37 d. Conclusion: The possibility of AI should be taken into account when the patients with AIDS have unexplained symptoms such as anorexia, nausea, weight loss and more characteristic manifestations such as orthostatic hypotension, hyponatremia or hyperkalemia.Early administration of glucocorticoid replacement therapy is feasible and effective.

Endoscopic submucosal dissection in treatment of esophageal granular cell tumor:A report of 2 cases and literature review

Objective: To report two cases of esophageal granular cell tumor treated by endoscopic submucosal dissection, and to improve the clinical awareness of the disease. Methods: The results of endoscope and ultrasonography endoscope of 2 cases were retrospectively analyzed. A preliminary diagnosis of the 2 cases was esophageal submucosal protruded lesion. The patients underwent endoscopic submucosal dissection and the tumors were removed completely. Firstly, the lesion border was marked and then hyaluronic acid was injected into submucosa to uplift the lesion sufficiently. Secondly, the mucosa was circumferentially incised along the marked points, and then the lesion was dissected along the submucosal layer gradually to the whole. Finally, necessary hemostatic was undertaken and the wound was clipped with titanium. Results: The pathological results after operation represented that the cells were polygonal or fusiform, rich in cytoplasm and had a dichromatic granular substance, and the nuclei were small.The immunohistochemical results showed S-100(+),NSE(+)/(-),CK(-),SMA(-),CD117(-),desmin(-) and proved that 2 cases of tumors were esophageal granular cell tumor. Two patients recovered smoothly after operation. They underwent endoscopic examination 16 months after operation, and no local recurrence or metastasis was founded. Conclusion: Endoscopic submucosal dissection is a safe and effective treatment of esophageal granular cell tumor,and the postoperative prognosis is good.

Evaluation on clinical effect of cast pure titanium split post crown restoration of molars with low occlusogingival distance and large root furcation

Objective: To observe the clinical effects of split post crown and split post-core crown in restoring the molars with low occlusogingival distance and large root furcation, and to clarify its advantages and analyze its improvement methods. Methods: Forty-nine molars of twenty-nine patients, which had been completely treated with root canal therapy, were restored by split post crown or split post-core crown according to their wills. Split post crown was used in twenty-three cases, and split post-core crown was applied in the other twenty-six cases. The clinical effects of the patients in two groups were observed after 12 and 24 months follow-up. The levels of MMP-8 in gingival crevicular fluid were checked and compared 12 months after restoration. Results: The restoration success rates of split post crown were 95.7% after 12-month treatment and 82.6% after 24-month treatment, respectively; there was no significant difference between them(P>0.05).The success rates of split post-core crown were 84.6% after 12-month treatment and 52.2% after 24-month treatment, respectively;there was significant difference between them(P<0.05). The success rate of split post crown was significantly higher than that of split post-core crown after 24-month treatment(P<0.05). The levels of MMP-8 in gingival crevicular fluid of the patients were close to each other, there was no statistical significance(P>0.05). Conclusion: The split post crown shows better clinical effect in restoring the molars with low occlusogingival distance in 24 months than split post-core crown. Targeted improvement will help to improve the clinical success rate.

Comparison of diagnostic values between magnetic susceptibility-weighted imaging and routine MRI in patients with cerebral amyloid angiopathy-related hemorrhage

Objective: To analyze the imaging features of magnetic susceptibility-weighted imaging (SWI) in the patients with cerebral amyloid angiopathy-related hemorrhage (CAAH), and to clarify the diagnostic value of SWI for CAAH. Methods: A total of 68 patients presumptively diagnosed as CAAH were collected,and their imaging data of routine MRI and SWI were collected and analyzed. The detection rates of hemorrhage focus of the CAAH patients were compared between two kinds of imaging examination. The consistency of detection of CAAH by routine MRI and SWI was analyzed. The imaging features of SWI and the risk of focus hemorrhage in the patients with CAAH were analyzed by multivariate Logistic regression analysis. Results: Sixty-one patients were confirmed as CAAH by pathologic diagnosis, and 53 patients were confirmed as CAAH by routine MRI; the detection rate was 86.89%; 59 cases of hemorrhage focus were confirmed by SWI and the detection rate was 96.72%. The number of lesions detected by SWI was more than that of routine MRI (P<0.05). The consistency of detection of CAAH by routine MRI and SWI was poor, and the value of Kappa was 0.3666. The patchy high signal and multiple clear edge low signal area were the relative imaging features of CAAH with SWI in the patients with CAAH analyzed by multivariate Logistic regression analysis (OR=3.895, P=0.025; OR=3.124, P=0.029). Conclusion: SWI can efficiently detect the hemorrhage focus in the patients with CAAH and the diagnostic value is better than routine MRI.

Analysis on prevalence of chronic obstructive pulmonary disease based on pulmonary function screening in Changchun urban area of Jilin province

Objective: To investigate the prevalence of chronic obstructive pulmonary disease (COPD) in the adults aged 40 years old and over in Changchun urban area, and to clarify the important significance of early application of pulmonary function examination. Methods: With the help of community workers,10 communities which around Second Hospital of Jilin University were selected and the participants aged 40 years old and over were randomly collected. COPD questionnaire and pulmonary function examination were conducted in all subjects. The patients with COPD were diagnosed according to the criteria established by the Global Initiative for COPD (GOLD). The patients with COPD were divided into the diagnostic group and undiagnostic group,then the patients with COPD who had smoking history were divided into smoking cessation group and smoking group.The pulmonary function of each groups were compared. Results: A total of 1 392 participants aged 40 years old and over were recruited in this survey, including 568 males and 824 females, and their average age was (59.59±6.60) years. Totally 176 subjects were diagnosed as COPD, and the prevalence was 12.64%. Furthermore, 160 of them were newly diagnosed, which showed the undiagnostic rate of COPD was 90.91%. Among the 176 patients with COPD, 16 had been confirmed before, which meant only 9.09% patients got the accurate diagnose before.The analysis of high-risk factor(smoking) showed that the population had an increased prevalence of COPD (20.71%) compared with non-smokers (8.8%)(P<0.05).The correlational analysis between smoking index and the pulmonary function of the patients with COPD showed that all of the pulmonary function indexes except forced vital capacity(FVC%) and maximum midexpiratory flow(MMEF%)in smoking cessation group were descended along with the increase of smoking index(P<0.05).All of the pulmonary function indexes except FVC% in smoking group were descended along with the increase of smoking index(P<0.05). Conclusion: The high prevalence and underdiagnostic rate of COPD are found in the residents aged 40 years old and over in Changchun urban area.More patients with COPD should be screened by performing pulmonary function examination to treat them at an early stage.

Investigation of depression status of hospitalized pregnant women in labor

Objective: To learn the depression status of the hospitalized pregnant women in labor and its related influencing factors, and to provide the theoretical basis for exploring depression intervention of the pregnant women in labor. Methods: Self-Rating Depression Scale was used for investigating the depression status of 203 hospitalized pregnant women in labor who were in hospital from November 2014 to May 2015, and the impacts of demographic characteristics, maternal history and physical condition and health care on depression status of hospitalized pregnant women in labor before pregnancy and during pregnancy were analyzed using Logistic regression. Results: Fifty-three people with depressive tendencies were detected among pregnant women in labor, and the positive rate was 26.1%. The differences in positive rates of depression tendency of the pregnant women between less than 30 years, 30-39 years and 40 years and above had no statistical significance (P>0.05). Univariate Logistic analysis showed that the differences of occupation, overweight or obesity, low-dose folic acid supplements before pregnancy were statistically significant (P<0.01). Multivariate Logistic regression analysis showed that risk factor for pregnant women was overweight or obesity pre-pregnancy, and the risk was 2.335 times to non-overweight or non-obesity pregnancy women (OR=2.335,95%CI:1.101-4.954,P<0.05). Conclusion: The pregnant women have the depression tendency, especially the pregnant women with high body mass index before pregnancy. The knowledge of psychological health education should be provided for them.
