Galectin-3及整合素ανβ3 mRNA在脑胶质瘤细胞中的表达及其意义

摘要:

目的:探讨半乳糖凝集素-3(Galectin-3)及整合素ανβ3 mRNA在胶质瘤细胞中的表达及二者在胶质瘤的发生及恶性进展中的作用。方法:采用原位杂交法检测5例正常脑组织及40例不同级别胶质瘤中Galectin-3 mRNA、整合素ανβ3 mRNA的表达情况,并分析二者之间的关系。结果:正常脑组织中Galectin-3 mRNA、整合素ανβ3 mRNA的表达均为阴性。Galectin-3 mRNA主要表达于脑胶质瘤细胞的胞浆中,在23例Ⅰ-Ⅱ级胶质瘤标本中阳性2例、弱阳性4例、阴性17例,阳性表达率为29.09%(6/23),阳性指数(LI)值为(4.80±3.22)%;在17例Ⅲ-Ⅳ级胶质瘤标本中强阳性4例、阳性6例、弱阳性1例、阴性6例,阳性表达率为64.71%(11/17),LI值为(27.30±22.72)%,Ⅰ-Ⅱ级胶质瘤Galectin-3 mRNA阳性表达率低于Ⅲ-Ⅳ级(P<0.05)。整合素ανβ3 mRNA主要表达于脑胶质瘤细胞的胞浆中,在23例Ⅰ-Ⅱ级胶质瘤标本中阳性3例、弱阳性4例、阴性16例,阳性表达率为34.43%(7/23),LI值为(5.35±3.79)%;在17例Ⅲ-Ⅳ级胶质瘤标本中强阳性4例、阳性6例、弱阳性3例、阴性4例,阳性表达率为76.47%(13/17),LI值为(29.24±21.10)%,Ⅰ-Ⅱ级胶质瘤整合素ανβ3 mRNA 阳性表达率低于Ⅲ-Ⅳ级(P<0.05)。直线相关分析结果显示,Galectin-3 mRNA与整合素ανβ3 mRNA的阳性细胞表达率呈正相关(r=0.718,P<0.05)。结论:Galectin-3 mRNA、整合素ανβ3 mRNA的表达在Ⅰ-Ⅱ级胶质瘤组明显低于Ⅲ-Ⅳ级胶质瘤组,提示Galectin-3及整合素ανβ3 mRNA检测可作为从分子水平判定人脑胶质瘤恶性生物学行为的指标。

Abstract:

Objective To study the expressions of galactose Galectin-3 and integrin ανβ3 mRNA in glioma cells  and their roles in the occurrence and the malignancy progress  of glioma.Methods  In situ hybridization was used to detect the expressions of Galectin-3 and integrin ανβ3 mRNA in 5 normal brain tissues and 40 patients with different grades gliomas , and the relationship between them was analyed. Results The expressions of Galectin-3 and integrin ανβ3 mRNA in normal brain tissue were negative.Galectin-3 mRNA mainly expressed in the cytoplasm and membrane of tumor cells.In 23 cases ofⅠ, Ⅱ grade glioma specimens, 2 cases were positive,4 cases were weakly positive and 17 cases were negative, the positive rate was 29.09%(6/23), LI value was (4.80±3.22)%;in  17 cases of Ⅲ, Ⅳ grade glioma specimens,4 cases were strongly positive,6 cases were positive ,1 case was weakly positive and 6 cases were negative,the positive rate was 64.71%(11/17), LI value was (27.30±22.72)%; there was significant difference in the positive rate betweenⅠ,Ⅱ grade and Ⅲ, Ⅳgrade glioma(P<0.05). The integrin ανβ3 mRNA mainly expressed in the cytoplasm and membrane of tumor cells. In 
23 cases of Ⅰ, Ⅱ grade glioma specimens 3 cases were positive expressin of integrin αυβ3,4 cases were weakly positive, 16 cases were negative, the positive rate was 34.43% (7 / 23), LI value was (5.35 ± 3.79)%; in  17 cases of Ⅲ, Ⅳ grade glioma specimens,4 cases were strongly positive, 6 cases were positive, 3 cases were weakly positive, and 4 cases were negative,the positive rate was 76.47% (13/17), and LI value was (29.24± 21.10)%; there was significant difference in the positive rate betweenⅠ,Ⅱ grade and Ⅲ, Ⅳgrade glioma(P<0.05).The linear correlation analysis showed that the expression rates of Galectin-3 and integrin ανβ3 mRNA  were positively correlated (P<0.05).Conclusion The expressions of Galectin-3 and integrin ανβ3 mRNA  in Ⅰ, Ⅱ grade gliomas are significantly lower than those of Ⅲ, Ⅳ grade glioma,which indicates that the detection of  Galectin-3 and integrin  ανβ3 mRNA  can be used as an indicator to judge malignant biological behavior of human brain glioma at molecular level.
