Cell cycle uncoupling indued by DNA damage

摘要: 目的:观察DNA损伤因子丝裂霉素 (MMC) 能否诱导细胞周期解偶联。 方法:采用PI荧光标记及流式细胞术 (FCM) 测定细胞周期并分析细胞倍体的变化。 结果:1.0及2.0 mg•L-1 MMC作用后24 h, SKOV-3细胞八倍体数显著增高(P<0.01, P<0.05)。1.0、2.0及4.0 mg•L-1 MMC作用后24 h,L929及EL-4细胞八倍体数显著增高(P<0.05,P<0.01,P<0.01及P<0.01,P<0.05,P<0.01)。结论:MMC可诱导人卵巢癌细胞SKOV-3、小鼠成纤维细胞L929及小鼠淋巴瘤细胞EL-4发生细胞周期解偶联。

Abstract: Objective To investigate whether cell uncoupling could be induced by mitomycin C (MMC). Methods Cell cycle was analyzed by flow cytometry (FCM) following staining of cells with propidium iodide (PI) and the changes of polyploid cells were observed.Results The 8N cells were increased significantly compared with control,24 hours after exposure of SKOV-3 cells to MMC with the doses of 1.0 and 2.0 mg•L-1 (P<0.01, P<0.05, respectively). And 24 hours after exposure of L929 cells and EL-4 cells to MMC with the doses of 1.0, 2.0, and 4.0 mg•L-1, the 8N cells were also increased significantly compared with control (P<0.05 or P<0.01). Conclusion The cell cycle uncoupling could be induced by MMC exposure.
