STEAP1在人前列腺组织中的表达与分布

摘要: 目的:探讨STEAP1在中国人前列腺组织中的表达和分布情况。方法:应用RT-PCR、免疫印迹和免疫组织化学染色的方法,检测STEAP1在正常前列腺、前列腺癌组织中mRNA和蛋白表达水平及其在细胞中的定位。 结果:RT-PCR结果显示前列腺癌STEAP1 mRNA高表达,正常前列腺则表达较弱;免疫印迹结果表明前列腺癌组织中STEAP1的表达水平高于正常前列腺组织的表达量;免疫组织化学染色显示阳性染色细胞主要分布在正常前列腺腺上皮细胞和前列腺癌腺上皮细胞,间质细胞、基底细胞染色阴性,着色部位主要在细胞膜及胞浆,而且在腺上皮细胞表现出一定的方向性, 主要分布在前列腺腺管管腔一侧。结论:STEAP1在中国人前列腺癌组织中广泛表达,主要表达于靠近前列腺腺管一侧的前列腺上皮细胞膜,依据结构与功能相结合的原则,其表达的解剖分布与表达水平提示STEAP1与前列腺内物质代谢有关。

Abstract: Objective To examine the expression and distribution of six transmembrane epithelial antigen of the prostate (STEAP1) in human prostate tissues. Methods The RT-PCR for mRNA, Western blotting for protein, immunohistochemistry for distribution were carried out. Results The results of RT-PCR and Western blotting showed that STEAP1 mRNA and protein highly expressed in prostate cancer tissues than those in normal prostate tissues. Immunohistochemistry staining showed that most staining positive cells were prostate epithelial cells , they located in the membrane of epithelial cells, and mainly in the side of cell membrane to the glandular vessels of the prostate.Conclusion STEAP1 express widely in human prostate cancer tissu es, and is closely related to epithelial cells, which is closely connected with the glandular of prostate. The expression and distribution indicate that STEAP1 is involved in substance metabolism and transportation in prostate.
