Effects of specific expression of α-catulin gene on proliferation and apoptosis of tongue squamous cell carcinoma TSCCA cells

Abstract:

Objective: To explore the effects of silencing expression of α-catulin gene on the proliferation and apoptosis of human tongue squamous cell carcinoma(TSCC) TSCCA cells,and to clarify its role in the biological behavior of TSCCA cells. Methods: The TSCCA cells at logarithmic growth phase were selected and divided into α-catulin-siRNA transfection group,empty plasmid transfection group, and blank control group.The relative expression levels of α-catulin mRNA in the TSCCA cells in various groups were tested by RT-PCR method;the relative expression levels of α-catulin protein in TSCCA cells in various groups were detecetd by Western blotting method;the inhibitory rate of proliferation of TSCCA cells was checked by MTT method;The apoptotic rates of TSCCA cells were measured by flow cytometry(FCM). Results: The RT-PCR results showed that the relative expression level of α-catulin mRNA in the TSCCA cells in α-catulin-siRNA transfection group was lower than those in empty plasmid transfection group and blank control group(P<0.01); the relative expression levels of α-catulin mRNA were decreased gradually with the prolongation of time,and the relative expression level of α-catulin mRNA at 72 h was lower than those at 24 and 48 h(P<0.01).The Western blotting results revealed that compared with empty plasmid transfection group and blank control group,the relative expression level of α-catulin protein in the TSCCA cells in α-catulin-siRNA transfection group was decreased(P<0.05).The MTT results demonstrated that the inhibitory rate of growth of TSCCA cells in α-catulin-siRNA transfection group was higher than those in empty plasmid transfection group and blank control group(P<0.001).The FCM results exhibited that the apoptotic rate of TSCCA cells in α-catulin-siRNA transfection group was increased compared with empty plasmid transfection group and blank control group(P<0.001). Conclusion: Silencing the expression of α-catulin RNA specifically could suppress the growth of TSCCA cells and promote the apoptosis of TSCCA cells.
