Screening of differentially expressed proteins in ovarian carcinoma tissue |and research on signaling pathway

Abstract:

Abstract:Objective  To detect the different proteins in ovarian carcinoma tissues and explore the relationship between the proteins and the carcinogenesis and development of ovarian carcinoma. Methods 19 cases of serous carcinoma tissues and 12 cases of normal ovarian tissues were collected, the proteins from tissues were extracted. With protein pathway array technique,37 kinds of phosphorylative antibodies and 73 kinds of  non-phosphorylative antibodies, the different proteins in ovarian carcinoma tissues and normal ovarian tissues were detected. The results were analyzed by QUANTITY One software and statistically calculated. Results There were 52 proteins expressed (47%) and 23 proteins (21%) were diferentially expressed with statistical significance (P<0.05), including the over expression proteins: p-Stat3, p-PDK1, p-cdc2, p-p38, p-AKT, XIAP, PCNA, cdc2p34, p53, Cdk2, Mesothelin, E-cadherin and down-regulated proteins: ERK, Cdc25C, Wee 1, Akt, N-cadherin, α-tubulin, Vimentin, cdc42, Bax, PTEN (P<0.05). Compared with normal ovarian tissues and normal ovarian cell line,p-Stat3, p-PKD1 and p-AKT were over expressed in ovarian carcinoma tissues and cell lines (P<0.05). Conclusion Stat3, PKD1 and AKT play important roles in carcinogenesis of ovarian carcinoma
