PRPS2表达特点及其与乳腺癌预后关系的生物信息学分析

Bioinformatic analysis on expression characteristics of PRPS2 and its relationship with prognosis of breast cancer

摘要: 目的

研究磷酸核糖焦磷酸合成酶2(PRPS2)在不同亚型乳腺癌中的表达及其与乳腺癌预后关系。

方法

采用GEPIA数据库研究泛乳腺癌和正常乳腺组织中PRPS2 mRNA表达水平;使用UALCAN数据库基于患者年龄、是否绝经、乳腺癌不同分子亚型、不同组织亚型、病理分级和是否存在淋巴结转移等层面分析PRPS2在乳腺癌和正常乳腺组织中的表达;采用miRTarBase数据库预测与PRPS2 3'UTR 结合的miRNAs。应用Kaplan-Meier plotter数据库研究与PRPS2和PRPS2 3'UTR 结合的miRNAs与乳腺癌预后的关系。

结果

与正常乳腺组织比较,在不同亚型乳腺癌组织中PRPS2 mRNA表达水平明显升高(P<0.05);PRPS2 mRNA表达水平越高,乳腺癌患者的生存期越短。与正常乳腺组织比较,在不同亚型乳腺癌中PRPS2 3'UTR结合的hsa-miR-215-5p和hsa-miR-26b-5p表达水平均明显降低(P<0.05),hsa-miR-215和hsa-miR-26b表达水平越低,乳腺癌患者的生存期越短。

结论

PRPS2具有泛乳腺癌特征性基因表达特点,乳腺癌中可能存在miR-PRPS2-乳腺癌预后的调控轴。

Abstract: Objective

To investigate the expressions of phosphoribosyl pyrophosphate synthetases 2 (PRPS2) in different subtypes of breast cancer and its relationship with the prognosis of breast cancer.

Methods

The transcription levels of PRPS2 in the pan-breast cancer and normal breast tissues were investigated using GEPIA database. Furthermore, UALCAN database was used to analyze the expressions of PRPS2 in the breast cancer and normal breast tissues on the basis of age, menopause status, different subtypes, histologic subtypes, pathological stages, and nodal metastasis status. miRTarBase database was used to predict the miRNAs binding with 3' un-translated region (3'UTR) of PRPS2. Kaplan-Meier plotter database was used to evaluate the relationships between the miRNAs binding with PRPS2 andPRPS23'UTR and the prognosis of breast cancer.

Results

Compared with normal breast tissue,the expression levels of PRPS2 mRNA in different subtypes of breast cancer tissue was significantly increased(P<0.05).The high expression of PRPS2 mRNA documented the poor prognosis of breast cancer. Compared with normal breast tissue,the expression levels of hsa-miR-215-5p and hsa-miR-26b-5p, the miRNAs binding with 3'UTR of PRPS2,in the different subtypes of breast cancer tissue were significantly reduced(P<0.05);moreover, the low expression levels of hsa-miR-215 and hsa-miR-26b reflected the poor prognosis of breast cancer.

Conclusion

PRPS2 is of specific expression characteristics of pan-breast cancer, suggesting that the axis of miR-PRPS2-prognosis of breast cancer may exist.
