MAOA基因多态性与内化性精神障碍的关联性分析

Association between MAOA gene polymorphism and internalizing disorders

摘要:

目的: 探讨MAOA基因多态性与内化性精神障碍的关系,为内化性精神障碍的易感基因研究奠定基础。方法:对194例内化性精神障碍大学生(病例组)和177名健康大学生(对照组)MAOA基因上的tag SNP位点采用多重聚合酶链式反应 (PCR) 和连接酶检测反应 (LDR) 的方法进行基因型检测。采用拟合优度χ2检验分析基因型频率分布是否符合Hardy-Weinberg平衡,应用χ2检验分析病例组与对照组,仅患1种内化性精神障碍组、患2种及2种以上内化性精神障碍组和对照组3组中该tag SNP位点的基因型和等位基因频数分布与内化性精神障碍的关联性。结果: 病例组和对照组中的rs2283724位点的基因型频率分布均符合Hardy-Weinberg 平衡定律 (χ2 =3.763,P>0.05;χ2 = 3.213,P>0.05)。rs2283724位点在病例组与对照组,仅患1种内化性精神障碍组、患2种及2种以上内化性精神障碍组和对照组3组中的基因型和等位基因频数分布差异均无显著性(P>0.05)。结论:MAOA基因可能与内化性精神障碍的发生无关联,提示MAOA基因可能不是内化性精神障碍发生的主要易感基因。

Abstract:

Abstract:Objective To investigate the association between MAOA gene polymorphism and internalizing disorders and provide theoretical basis for its susceptible gene study.Methods The method of PCR-based ligase detection reaction (PCR-LDR) was applied to detect the genotypes of the tag SNP on MAOA gene in 194 cases affected by internalizing disorders and 177 controls of healthy undergraduates.Hardy-Weinberg equilibrium for genotypic distribution was estimated by the goodness of fit χ2 test.The distribution of allelic and genotypic frequencies of MAOA gene among case groups,control group and affected different number of internalizing disorders groups was statistically computed.Results The genotypic frequency of MAOA gene did not deviate from Hardy-Weinberg equilibrium in both case and control groups (χ2 =3.763,P>0.05;χ2 = 3.213,P>0.05).The tag SNP was not associated with the disorders (P>0.05).There were no correlations between allelic or genotypic frequencies among the different degrees of co-morbidities and control group (P>0.05). Conclusion MAOA gene may not be associated with internalizing disorders,which means MAOA gene may not be the major gene caused the disease.
