caspase-8在胶质母细胞瘤抵抗TRAIL诱导凋亡中的作用

Role of caspase-8 in glioblastoma resistance to TRAIL-induced apoptosis

摘要:

目的:对人脑胶质母细胞瘤SC189细胞株中单克隆细胞株进行研究,探讨caspase-8在胶质母细胞瘤抵抗肿瘤坏死因子相关凋亡诱导配体(TRAIL)诱导凋亡分子机制中的作用。方法:获取SC189单克隆细胞株,应用酸性磷酸酶法检测SC189单克隆细胞株对TRAIL敏感性;Western blotting检测各单克隆细胞株表达FADD、DR5、DR4及caspase-8情况;经TRAIL作用后发生caspase-8、caspase-3、caspase-9、DFF-45凋亡级联裂解反应情况。结果:获得的5株SC189单克隆细胞株,经不同浓度TRAIL作用后细胞死亡率为-5.25%~45.80%,其中4株对TRAIL抵抗,1株对TRAIL相对敏感;Western blotting检测发现FADD、DR5、DR4表达相似,caspase-8表达不同,发生抵抗的细胞株caspase-8表达明显减少,经TRAIL作用后不发生凋亡级联裂解反应;相对敏感的细胞株caspase-8表达增多,经TRAIL作用后可出现明显的凋亡级联裂解反应。结论:SC189单克隆细胞株中caspase-8表达量直接与TRAIL反应敏感性有关联。

Abstract:

Abstract:Objective
To study the monoclones of SC189 and discuss the role of caspase-8 in glioblastoma resistance to tumor necrosis factor related apoptosis-inducing ligand(TRAIL)-induced apoptosis.Methods The monoclones were obtained from SC189,then the acid phosphate assay was used to detect the cell death rate to TRAIL,the expression of FADD,DR5,DR4 and caspase-8 in monoclones were detected by Western blotting,and then the caspases cleavage was checked after treated with TRAIL.Results The death percentage of cells of the monoclones isolated from SC189 was -5.25%-45.8% after treated with different doses of TRAIL,4 monoclones were resistant cell lines,only one monoclone was sensitive to TRAIL.Western blotting detection showed that the expressrons of FADD,DR5 and DR4 were similar.The expression of caspase-8  in resistant cell lines was decreased,and the cells hadn’t the cleavage reaction,otherwise,the expression of caspase-8 in sensitive cell lines was increased,and the cells had the caspases and DFF-45 cleavage after treated with TRAIL.Conclusion The expression of caspase-8 in SC189 monoclones is directly related to the sensitivity of TRAIL.
