Expression characteristics of Wnt5a in different periods of cementum development and its effect on differentiationof cementoblasts of mice

Objective: To study the expression characteristics of Wnt5a in cementum of the mice after birth and the effect of Wnt5a on the differentiation of cementoblasts in vitro,and to illuminate the mechanism of Wnt noncanonical signaling in regulating the tooth development. Methods: The postnatal Kunming mice at the days 0.5,4.5,12.5,18.5,24.5 and 30.5 after birth were selected and divided into various groups by different time points (n=3).The mice were sacrificed and the lower first molar and peridentium were prepared.Immunohistochemical staining was used to detect the expression characteristics of Wnt5a in tooth and peridentium of the mice.The OCCM30 cells were divided into control group and experiment group.200 μg·L-1 recombinant Wnt5a protein were added in experiment group, and nothing was added in control group;the total RNA in the cells was exrracted.The relative expression levels of Ocn,Opn,Alp,Bsp,Wnt5a,and Runx-2 genes in the OCCM30 cells in two groups were detected by RT-PCR method. Results: At the day 0.5 after birth,the expression of Wnt5a in the lower first molar tooth germ was negative.At the day 4.5 after birth,the expression of Wnt5a in ameloblasts and odontoblasts were positive.At the day 16.5 after birth,the expression of Wnt5a in cementoblasts was positive.At the days 20.5-30.5 after birth,the expressions of Wnt5a in odontoblasts and periodontal ligament cells were positive.Compared with control group,the relative expression levels of Ocn and Alp gene in the OCCM30 cells in experiment group were decreased (P<0.05). Conclusion: The expression of Wnt5a is regular at different tooth developmental stages after birth.Wnt5a can reduce the expression of differentiated genes and play an important role in the growth of cementum.

Inhibitory effect of scutellarin on proliferation of human glioma U87 cellsand its mechanism

Objective: To explore the inhibitory effect of scutellarin(SCU) on the proliferation of human glioma U87 cells,and to clarify its mechanism. Methods: The U87 cells were cultured and divided into blank control group and 20,40,80 mg·L-1 SCU groups,the surival rates of U87 cells in various groups were examined by MTT assay; the morphology of U87 cells in various groups were observed by inverted microscope;the apoptotic rates of U87 cells were examined by Annexin Ⅴ/PI staining; the expression levels of bax and bcl-2 proteins in the U87 cells in various groups were detected by Western blotting method. Results: The MTT results showed that compared with blank control group,the survival rates of U87 cells in 20,40,and 80 mg·L-1 SCU groups were decreased(P<0.01).The inverted microscope observation results showed that compared with blank control group,the number of U87 cells in 80 mg·L-1 SCU group was significantly decreased,and the cell volume was smaller;the U87 cells were shrinkage,shedding,and the outline of adherent cells was fuzzy.The results of AnnexinⅤ/PI staining showed that the late and total apoptotic rates of U87 cells in 80 mg·L-1 SCU group were increased compared with blank control group (P<0.01).The Western blotting results showed that compared with blank control group,the expression levels of bcl-2 protein in the U87 cells in 20,40 and 80 mg·L-1 SCU groups were decreased,especially in 80 mg·L-1 SCU group(P<0.01).Compared with blank control group,the ratio of bax/bcl-2 in the U87 cells in 80 mg·L-1 SCU group was significantly increased(P<0.01). Conclusion: SCU can induce the apoptosis and suppress the tumor growth by upregulating the bax/bcl-2 ratio in the U87 cells.

Enhancement of histone deacetylase inhibitors SAHA in chemotherapeutic sensitivity of ovarian cancer SKOV3 cells

Objective: To investigate the biological effects of histone deacetylase inhibitors(HDACIs) suberoylanilide hydroxamic acid(SAHA) combined with vincristine (VCR) on the ovarian cancer SKOV3 cells, and to clarify their mechanisms. Methods: The human ovarian cancer SKOV3 cells were divided into control group,SAHA group,VCR group and SAHA+VCR group.CCK-8 method was used to detect the surival rates of SKOV3 cells in various groups.The apoptotic rates,the autophagy rates and the distribution of cell cycles of SKVO3 cells in various groups were detected by flow cytometry (FCM). Results: The CCK-8 results showed that compared with control group,the survival rates of SKOV3 cells in SAHA group and VCR group were decreased significantly.Compared with VCR group or SAHA group,the survival rates of cells in SAHA+VCR groups were decreasedsignificantly(P<0.05).The FCM results demonstrated that compared with control group, the apoptotic rates,the autophagy rates and the percentages of the SKOV3 cells at G2/M phase in SAHA group and VCR group were increased(P<0.05);compared with SAHA or VCR groups, the apoptotic rate,the autophagy rate and the percentages of SKOV3 cells at G2/M phase in SAHA+VCR groups were increased(P<0.05). Conclusion: SAHA can enhance the sensitivity of ovarian cancer SKOV3 cells to VCR and induce the apoptosis and autophagy of SKOV3 cells to promote the cell arrest at G2/M phase.The combined application of SAHA and chemotherapeutic drugs may be a new strategy for the treatment of ovarian cancer patients.

Effect of hyperoxia on NRP-cGMP signaling pathway in brain tissue of newborn rats

Objective: To construct the brain tissue damage models of newborn rats induced by hyperoxia,and to explore the effect of hyperoxia on the natriuretic peptide receptor-cyclic guanosine monophosphate(NRP-cGMP) signaling pathway in brain tissue of the newborn rats. Methods: A total of 60 newly born Wistar rats were divided into control group and hyperoxia model group,and there were 30 rats in each group. At the 1st,3rd and 7th days after hyperoxia exposure,the brain tissues of 10 rats randomly selected from each group were gotten,and the body weights of the rats in two groups were detected.HE staining was used to observe the morphology of brain tissue of the rats in two groups;the ultrastructures of brain tissue of the rats in two groups were observed under transmission electron microscope.Western blotting method was used to detect the expression levels of natriuretic peptide receptor A (NPR-A) and natriuretic peptide receptor B (NPR-B) in brain tissue of the rats in two groups.ELISA method was used to determine the levels of cGMP in brain tissue of the rats in two groups. Results: The body weight of the rats in hyperoxia model group was significantly lower than that in control group at the 1st,3rd and 7th days after hyperoxia exposure(P<0.05).The HE staining results showed that at the 3rd day after hyperoxia exposure,the volume of the hippocampal pyramidal cells of the rats in hyperoxia model group was reduced, and the arrangement was sparse;at the 7th day after hyperoxia exposure,the hippocampal cells showed deep staining and shrinkage, and the cell boundaries were not clear. Under electron microscope,at the 3rd day after hyperoxia exposure, the mitochondrial double membrane structure of the rats in hyperoxia model group was destroyed,a few crest disappeared,and the number of mitochondria and synapses was decreased; the above damage conditions were aggravated at the 7th day after birth.The Western blotting results showed that at the 1st,3rd and 7th days after hyperoxia exposure,the expression level of NPR-A in brain tissue of the rats in hyperoxia model group was higher than that in control group(P<0.05).But only at the 1st day after hyperoxia exposure,the expression level of NPR-B in brain tissue of the rats in hyperoxia model group was higher than that in control group(P<0.05).The ELASA results showed that at the 1st,3rd and 7th days after hyperoxia exposure, the level of cGMP in brain tissue of the rats in hyperoxia model group was higher than that in control group(P<0.05). Conclusion: NRP-cGMP signaling pathway is involved in the occurrence of brain injury induced by hyperoxia.

Regulation effect of F-actin on senescence of mesenchymal stem cells and its mechanism

Objective: To investigate the regulation effects of fibrous-actin (F-actin) on senescent of the human bone marrow mesenchymal stem cells (hBMSCs), and to elucidate the molecular mechanisms of senescence the of hBMSCs. Methods: The hBMSCs were cultured in vitro and divided into control group (P2 hBMSCs), F-actin inhibitor group (P2 hBMSCs were treated with 2.5 μmol·L-1 F-actin inhibitor Latrunculin B for 1 h) and P11 hBMSCs group (P11 hBMSCs passaged from P2 hBMSCs continuously). The hBMSCs in various groups were induced by osteogenic, adipogenic and chondrogenic induction mediums and the induction effects were identified by Alizarin red staining, SO staining and oil red O staining. The number of Ki67 positive cells, polymerization and morphology of F-actin and YAP subcellular localization were detected by immunofluorescence staining. SA-β-Gal staining was uesd to detect the SA-β-Gal staining positive cells in the hBMSCs in various groups. Results: The Alizarin red staining, SO staining and oil red O staining results showed that the hBMSCs in various groups had the osteogenic, adipogenic and chondrogenic abilities.The immunofluorescence staining and SA-β-Gal staining results showed that the microfilament bundles in the hBMSCs in control group were more thick, the F-actin length was longer, and the YAP mainly concentrated in the nucleus (YAP was activated in the nucleus). Compared with control group, the number of Ki67 positive cells in P11 hBMSCs group was less and the number of SA-β-Gal positive cells was more;the Factin was shorter and thinner,the YAP mainly concentrated in the cytoplasm,and the high cytoplasmic YAP cells was positive for SA-β-Gal staining;in F-actin inhibitor group,the high cytoplasmic YAP inactivation was found in the hBMSCs and the SA-β-Gal staining positive hBMSCs had more senescent cells. Conclusion: To inhibit the entering of YAP into the nucleus can promote the senescence of hBMSCs and F-actin can affect the senescence of hBMSCs by regulating the YAP activity.

Preventive effects of total coumarin and essential oil of Angelicae Dahuricae Radix on migraine rats and their mechanisms

Objective: To observe the preventive effects of combined application of total coumarin and essential oil of Angelicae Dahuricae Radix on the migraine rats induced by nitroglycerin,and to explore their mechanisms. Methods: A total of 56 Wistar rats were randomly divided into normal control group (saline 0.1 mL·10 g-1),migraine model group (saline 0.1 mL·10 g-1),total coumarin of Angelicae Dahuricae Radix group(100 mg·kg-1),essential oil of Angelicae Dahuricae Radix group(100 mg·kg-1),and low dose (25 mg·kg-1),middle dose(50 mg·kg-1)and high dose(100 mg·kg-1)of total coumarin and essential oil composition of Angelicae Dahuricae Radix groups(composition groups)(n=8).After continuously intragastric adminstration for 7 d,the rats were injected subcutaneously with nitroglycerin (10 mg·kg-1) for establishing the migraine models.The behavior of the rats were observed,the levels of nitric oxide (NO) in serum and brain tissue of rats,the levels of calcitonin gene related peptide (CGRP) and endothelin (ET) in plasma of the rats in various groups were detected. Results: Compared with normal control group,the frequencies of head shaking,the face shoting times of posterior legs and the scratching times of anterior limbs of the rats in migraine model group were increased(P<0.05 or P<0.01),and the levels of NO in serum and brain tissue of the rats were increased(P<0.01).Compared with migraine model group,the frequencies of head shaking,the face shoting times of posterior legs and scratching times of anterior limbs of the rats in total coumarin group,essential oil group, and composition groups were decreased (P<0.05 or P<0.01),and the levels of NO in serum and brain tissue of the rats were decreased (P<0.05 or P<0.01). Compared with migraine model group,the levels of CGRP and ET in plasma of the rats in essential oil group,total coumarin group and middle and high doses of composition groups were decreased(P<0.05). Conclusion: The composition of two active ingredients of Angelicae Dahuricae Radix, total coumarin and essential oil, has preventive effects in the migraine rats induced by nitroglycerin,and their mechanisms may be related to regulation of the levels and function of vasoactive substances.

Effect of herba artemisiae capillaris extracts on expression profile of miRNAs in kidney tissue of diabetic rats and its protective effect on kidney

Objective: To observe the effect of herba artemisiae capillaris extracts (HACE) on the expression profile of microRNA (miRNAs) in the kidney tissue of the diabetic rats,and to explore the protective effect of HACE on the kidney tissue of diabetic rats from the miRNA angle. Methods: A total of 60 male Wistar rats were divided into control group (n=12),model group (n=24) and HACE group (n=24).The urinary albumin excretion rate and urinary protein excretion rate of the rats in various groups were detected. HE staining was used to detect the morphology of the kidney tissue of the rats in various groups after treated for 8 and 16 weeks; miRNAs chips were used to screen the expressions of miRNAs in the kidney tissue of the rats.The positive miRNAs were screened out from those with differentially expression amounts higher than 1.74 times as well as high abundance,and RT-qPCR was used to verify the relative expression amounts of miRNAs. Results: Compared with model group,the glomeruli matrix accumulation of the rats in HACE group(after treated for 8 and 16 weeks) was significantly improved and the urinary albumin excretion rates were decreased(P<0.05).The miRNAs chips results showed that there were 35 differentially expressed miRNAs in control and model groups and there were 17 differentially expressed miRNAs in model and HACE groups. The RT-qPCR results showed that after treated for 8 weeks, compared with control group,the relative expression amounts of miR-1306-3p(P<0.05),miR-672-5p(P<0.05) and miR-3550(P<0.01) in the kidney tissue of the rats in model group were decreased;compared with model group,the relative expression amount of miR-672-5p in the kidney tissue of the rats in HACE group was increased (P<0.05).After treated for 16 weeks,compared with control group,the relative expression amount of miR-21-5p in the kidney tissue of the rats in model group was increased(P<0.01),the relative expression amounts of miR-1306-3p(P<0.05),miR-672-5p(P<0.01) and miR-466d(P<0.05) were decreased;compared with model group,the relative expression amounts of miR-21-5p(P<0.05) and miR-1306-3p(P<0.05) in the kidney tissue of the rats in HACE group were decreased,and the relative expression amount of miR-672-5p was increased(P<0.05). Conclusion: The miRNAs expression profile in kidney tissue of diabetic rats were changed after HACE treatment.HACE has the protectively effect on the kidney tissue of diabetic rats and the mechanism may be associated with miRNAs.

Dynamic pathological characteristics of kidney in db/db mice with type 2 diabetes mellitus

Objective: To investigate the dynamic pathological characteristics of kidney in the db/db mice with type 2 diabetes mellitus and to clarify the pathogenesis and development mechanism of diabetic nephropathy,and to provide the experimental evidence for the further study on diabetic nephropathy. Methods: The male SPF db/db mice aged 7-8 weeks were selected as model group (n=16) and the db/m mice with the same ages were selected as control group(n=16).The body weights and levels of fasting blood glucose(FBG) of the mice were detected after 8,16, and 32 weeks.Then eight mice in each group were sacrificed and the kidney tissue was dissected at 8,16 and 32 weeks.HE and Masson staining were used to observe the pathomorphology of the kidney tissue; the ultrastructures of kidney tissue were observed under electron microscope. Results: Compared with normal group,the body weights of the mice in model group at 8,16 and 32 weeks were significantly increased(P<0.01),the FBG levels were significantly increased(P<0.01),and the double kidney indexes were significantly decreased(P<0.01);the double kidney index of the mice in model group at 32 weeks was lower than that at 16 weeks(P<0.05).The HE and Masson staining results showed that the morphological changes of kidney tissue of the mice at 16 weeks were significant,the glomerular volume was expansion and the kidney tubular epithelial cells were edema,which turned serious at 32 weeks.A large number of blue dye substances in the kidney tissue was found.Under electron microscope,the glomerular basement membranes got thickened,the foot processes got fused and the number of mitochondria in kidney tubular epithelial cells was reduced and swell was found in the mice in model group at 16 weeks;the lesions in the kidney tissue were serious at 32 weeks,and the collagenous fiber was visible at 32 weeks. Conclusion: The pathological changes in the kidney tissue of the db/db mice at 16 weeks are significant,such as glomerular basement thickening and footwork fusion.The kidney tissue of the db/db mice at 32 weeks shows prominent fibrosis.

Repair effect of dental pulp stem cells on facial nerve injury in rabbits and its mechanism

Objective: To establish the rabbit facial nerve injury model by surgery and to explore the repair effect of dental pulp stem cells on the facial nerve injury in the rabbits,and to clarify its possible mechanism. Methods: The dental pulp stem cells were isolated,cultured and induced.A total of 45 rabbits were randomly divided into normal control group,model group and experiment group,and there were 15 rats in each group.Except for normal control group,the rabbits in the other groups were cut along the corner of mouth to the front of the ear to form a incision about 3 cm,the upper buccal branches were disconnected,and the facial nerve buccal injury models were established.After 1 week,0.1 mL dental pulp stem cell suspension (5×106) was injected into the operative cavity of the rats in experiment group, and the rabbits in normal control group did not receive any treatment.The rabbits in model group were injected with the same amount of phosphate buffer (PBS).Two weeks after operation,the behavior and movement function scores of facial beard of the rabbits in various groups were evaluated;the pathomorphology of facial nerve tissue of the rabbits in various groups was observed by HE staining;immunohistochemical staining was used to detect the number of brain derived nerve growth factor (BDNF) and ciliary neurotrophic factor (CNTF) positive cells; the number of regenerated nerve fibers,the diameter of fiber and the thickness of myelin sheath were observed under transmission electron microscope. Results: The primary dental pulp stem cells of young rabbits were successfully separated and identified.Most of them were fibroblast like and long fusiform.The HE staining results showed that the nucleus of the third generation of the dental pulp stem cells showed a dark blue and oval shape,and the spindle shaped hyperchromatic cells were fibroblast like cells.Compared with normal control group,the movement function score of facial beard of the rabbits in model group was significantly decreased(P< 0.05);compared with model group,the movement function score of facial beard of the rabbits in experiment group was increased (P<0.05).Compared with model group,after treated for 10 weeks,there were more regenerated nerve fibers which were fascicular and tight.Compared with normal control group,the number of BDNF and CNTF positive cells in facial nerve tissue section of the rabbits in model group was decressed (P< 0.05);compared with model group,the number of BDNF and CNTF positive cells in facial nerve tissue section of the rabbits in experiment group was significantly increased (P<0.05). Compared with normal control group,the number of regenerated nerve fibers,the diameter of fiber and the thickness of myelin sheath of the rabbits in model group were significantly decreased(P<0.05);compared with model group,the number of regenerated nerve fibers,the diameter of fiber and the thickness of myelin sheath of the rabbits in experiment group were significantly increased (P<0.05). Conclusion: The dental pulp stem cells have a certain repair effect on the facial nerve injury,and the mechanism may be related to the up-regulation of the experessions of BDNF and CNTF.

Effect of SHH signaling pathway on apoptosis and invasion of trophoblast cells in preeclampsia patientsand its mechanism

Objective: To investigate the effect of activation of SHH signaling pathway on the apoptosis and invasion of the trophoblast cells in the preeclampsia(PE) patients,and to clarify its mechanism. Methods: The HTR8/SVneo cells were divided into normal control group,hypoxia/reoxygenation(H/R) treatment group and purmorphamine+H/R treatment group. The expression levels of SHH signaling pathway related proteins SHH,Ptch1,Smo,Gli1,and Gli3 in the placenta tissues of PE and late trimester of normal pregnancy were detected by Western blotting method;the apoptotic rates of HTR8/SVneo cells in various groups were detected by flow cytometry(FCM); Transwell assay was used to detect the number of transmembrane cells; the expression levels of matrix metalloproteinase-2(MMP-2) and metalloproteinase-9(MMP-9) proteins in the HTR8/SVneo cells in various groups were detected by Western blotting method. Results: The expression levels of SHH,Ptch1,Smo,Gli1 and Gli3 proteins in the placenta tissue of PE were significantly lower than those in normal pregnancy placenta tissue (P<0.05);the expression levels of SHH,Ptch1,Smo,Gli1,Gli3,MMP-2,and MMP-9 proteins and the number of transmembrane cells in H/R treatment group were significantly lower than those in normal control group(P<0.05),and the apoptotic rate was significantly higher than that in normal control group (P<0.05); the expression levels of SHH,Ptch1,Smo,Gli1,Gli3,MMP-2,and MMP-9 proteins and the number of transmembrane cells in purmorphamine+H/R treatment group were significantly higher than those in H/R treatment group(P<0.05),and the apoptotic rate was significantly lower than that in H/R treatment group (P<0.05). Conclusion: The activation of SHH signaling pathway can reduce the apoptosis of HTR8/SVneo cells in the placental tissue of the PE patients and promote the cell invasion by up-regulating the expressions of MMP-2 and MMP-9.

Promotion effect of endocrine gland-derived vascular endothelial growth factor on proliferation and migration of colon cancer LoVo cells

Objective: To investigate the effect of endocrine gland-derived vascular endothelial growth factor(EG-VEGF) on the malignant behavior of colon cancer LoVo cells. Methods: The LoVo cells treated with different concentrations (50,100,and 200 μg·L-1)of EG-VEGF were used as EG-VEGF groups,and the colon cancer LoVo cells cultured with solution without EG-VEGF were regarded as control group. The proliferation activity of LoVo cells was determined by MTT method; flow cytometry(FCM) was used to detect the percentages of LoVo cells at different cell cycles in various groups; Wound scratch assay test and Transwell cell migration assay were used to detect the migration rates of LoVo cells and the number of migration cells. Results: The MTT results showed that compared with control group,the proliferation activities of LoVo cells in 50,100,and 200 μg·L-1EG-VEGF groups were increased significantly (P<0.05); with the increasing of concentration of EG-VEGF,the proliferation activity was increased apparently. The FCM results showed that compared with control group,the percentage of colon cancer LoVo cells at G0/G1 phase in 100 μg·L-1EG-VEGF group was decreased (P<0.05) and the percentage of colon cancer LoVo cells at S phase was increased (P<0.05),but the percentage of colon cancer LoVo cells at G2+M phase did not change significantly.The wound scratch assay results showed that compared with control group,the migration rate of LoVo cells in 100 μg·L-1EG-VEGF group was increased (P<0.05). The Transwell assay results showed that compared with control group,the number of migration cells in 100 μg·L-1 EG-VEGF group was increased significantly (P<0.05). Conclusion: EG-VEGF can obviously promote the proliferation and migration of colon cancer LoVo cells,and EG-VEGF may be associated with the malignant development of colon cancer.

Effects of silencing expression of CRAF gene on invasion and metastasis of hepatocellular carcinoma cells

Objective: To investigate the effects of high expression of CRAF on the invasion and metastasis of hepatocellular carcinoma cells(HCC),and to clarify their mechanisms. Methods: The SMMC7721 cells with high expression of CRAF were regarded as the object,and the expressions of CRAF in SMMC7721 cells were down-regulated by shRNA.The SMMC7721 cells were divided into control group and silencing groups(shCRAF#1 transfection group and shCRAF#2 transfection group).The wound closure index of SMMC7721 cells was detected by cell scratch test,the number of transmembrane cells of SMMC7721 cells was detected by Transwell assay,and the expression levels of matrix metalloproteinase-2(MMP-2) and matrix metalloproteinase-9(MMP-9) in SMMC7721 cells were detected by Western blotting method. Results: The scratch test results showed that compared with control group,the wound closure indexes of the SMMC7721 cells in silencing groups were decreased (P<0.05).The Transwell assay results show that compared with control group,the number of transmembrane cells in silencing groups was decreased (P<0.05).The Western blotting results showed that compared with control group,the expression levels of MMP-2 and MMP-9 in the SMMC7721 cells in silencing group were decreased (P<0.05). Conclusion: Silencing CRAF can inhibit the invasion and migration of HCC SMMC7721 cells by inhibiting the expressions of MMP-2 and MMP-9.

Optimal extraction of Phaeoporus obliquus saponins and its scavengingeffect on fat accumulation in HepG2 cells

Objective: To investigate the ability of Phaeoporus obliquus saponins in scavenging the fat accumulation of liver cells,and to clarify its effect on the prevention of non-alcoholic fatty liver disease(NAFLD). Methods: The safe concentrations of Phaeoporus obliquus in the HepG2 cells was selected by MTT method. The cells treated with 60.000,6.000,0.600,0.060,and 0.006 g·L-1 Phaeoporus obliquus saponins were regarded as treatment groups;the blank well without drugs and cells was regarded as control group,and the HepG2 cells without drugs were regarded as model group.The survival rates of HepG2 cells in various groups were detected by MTT method;the clearance rates of triglyceride(TG) in the HepG2 cells in various groups were detected;the morphology of HepG2 cells was detected by oil red O staining. Results: The optimum extraction condition of Phaeoporus obliquus saponins were as followed:the liquid ratio was 1:15,the extraction temperature was 30℃,the extraction time was 30 min, and the ultrasonic power was 50%.Compared with control group,the survival rate of HepG2 cells in 0.060 g·L-1 Phaeoporus obliquus saponins group was decreased and the clearance rate of TG was increased(P<0.05),and the lipid interfusion in cells was significantly reduced.Compared with control group,the survival rate of HepG2 cells in 0.600 g·L-1 Phaeoporus obliquus saponins group was decreased and the clearance rate of TG was increased(P<0.01),and the number of lipid drops was significantly reduced. Conclusion: The extraction rate of Phaeoporus obliquus saponins is obviously improved under the optimum extraction conditions and it has good in vitro lipid-lowering activity,and it has better preventive effect on NAFLD.

Effect of melatonin on radiosensitivity of non-small cell lung cancer H1299 cells

Objective: To detect the effects of melatonin (MLT) combined with radiotherapy on the proliferation and apoptosis of non-small cell lung cancer (NSCLC) H1299 cells,and to explore the role of MLT in regulating the radiosensitivity of H1299 cells. Methods: The H1299 cells were divided into control group,MLT group,irradiation(IR) group and IR+MLT group. The H1299 cells in control group didn't receive any treatment, the H1299 cells in MLT group were treated with different concentrations of MLT (100 and 500 μmol·L-1),the H1299 cells in IR group were exposed to 6 Gy 137Cs γ-ray,and the H1299 cells in IR+MLT group were treated with different concentrations of MLT (100 and 500 μmol·L-1) and exposed to 6 Gy 137Cs γ-ray. Colony formation assay was used to detect the clone numbers of H1299 cells in various groups; Flow cytometry was applied to detect the apoptotic rates of H1299 cells at 24 and 48 h after irradiation; Western blotting method was performed to detect the phosphorylation levels of nuclear factor kappa B (NF-κB) protein in the H1299 cells in various groups. Results: The colony formation assay results showed that compared with IR group,the clone number of H1299 cells in IR+MLT group was significantly decreased(t=7.234,P<0.01).The flow cytometry results showed that the apoptotic rates of H1299 cells in IR group and MLT group at 24 h after irradiation were higher than that in control group (t=5.020,t=13.525,P<0.01); the apoptotic rate of H1299 cells in IR+MLT group at 24 h after irradiation was higher than that in IR group (t=12.884,P<0.01).There was no significant difference in the apoptotic rate of the H1299 cells between IR+MLT group and IR group at 48 h after irradiation(t=0.394,P>0.05).The Western blotting results showed that the phosphorylation level of NF-κB protein in the H1299 cells in IR+MLT group was lower than that in IR group. Conclusion: MLT combined with radiotherapy has obviously inhibitory effect on the human H1299 cells and can increase the radiosensitivity of H1299 cells.

Effects of CYP3A5 genetic polymorphisms on metabolism of tacrolimus in patients after kidney transplantation

Objective: To investigate the effects of different CYP3A5 genotypes on the concentrations of tacrolimus in blood of the patients after kidney transplantation,and to provide the valuable evidence for personalized administration in the patients after kidney transplantation. Methods: The clinical materials of 115 patients after kidney transplantation were analyzed retrospectively.The patients were divided into three groups according to the CYP3A5 genotypes:CYP3A5*1/*1 group(11 cases), CYP3A5*1/*3 group(46 cases) and CYP3A5*3/*3 group(58 cases).The concentration/dosage (C0/D) values of tacrolimus in blood of the patients in three groups at 7 d,1 month,3 months,6 months,1 year, and 2 years after operation were detected,respectively. Results: The differences of body mass index(BMI),age composition ratios and gender composition ratios of the patients were not significant between three groups(P>0.05).Compared with CYP3A5*1/*3 group,the C0/D values of tacrolimus in blood of the patients in CYP3A5*1/*1 group at 7 d,3 months,1 year and 2 years after operation were decreased(P=0.011,P<0.01,P=0.022,P=0.024);compared with CYP3A5*3/*3 group,the C0/D values of tacrolimus in blood of the patients in CYP3A5*1/*1 group and CYP3A5*1/*3 group at 7 d, 1 months,3 months,6 months,1 year and 2 years after operation were decreased(P<0.01).In CYP3A5*1/*1 group,the C0/D value of tacrolimus in blood of the patients at 7 d after operation was significantly lower than those at 6 months,1 year and 2 years after operation (P=0.004,P=0.049,P=0.036); in CYP3A5*1/*3 group,the C0/D value of tacrolimus in blood of the patients at 7 d after operation was significantly lower than those at 1 month,3 months,6 months,1 year and 2 years after operation(P=0.006,P<0.01,P<0.01,P<0.01,P<0.01),and the C0/D value of tacrolimus in blood of the patients at 1 month after operation was significantly lower than that at 6 months after operation(P=0.013); in CYP3A5*3/*3 group,the C0/D value of tacrolimus in blood of the patients at 7 d after operation was significantly lower than those at 1 month,3 months,6 months,1 year and 2 years after operation(P=0.002,P<0.01,P<0.01,P<0.01,P<0.01). Conclusion: The CYP3A5 genetic polymorphism has a significant impact on the metabolism of tacrolimus in blood of the patients after kidney transplantation,and the impact could last for two years.

Relationships between expressions of B7-H1 and B7-H4 and Foxp3+ regulated T-cell infiltration in colorectal cancer tissue

Objective: To explore the expressions of B7-H1 and B7-H4 in colorectal cancer(CRC) tissue and their relationships with the Foxp3+ regulated T-cell (Treg) infiltrationin tumor tissue,and to provide the basis for judging the biological behaviours and prognosis of CRC. Methods: Immunohistochemistry was used to detect the expressions of B7-H1 and B7-H4 and the infiltration of Treg in 56 cases of CRC tissue samples and adjacent normal tissue samples,and their relationships with the clinicopathological features of CRC patients were analyzed. Results: The positive expression rate of B7-H1 in CRC tissue of the CRC patients was higher than that in adjacent normal tissue(χ2=72.34,P<0.01).The differences of expression frequencies of B7-H1 in CRC tissues with different infiltration depths were significant (P<0.01). The high expression frequency of B7-H1 in CRC tissue of the patients with positive lymphnode metastasis was higher than that in the patients with negative lymphnode metastasis(P<0.01).The high expression frequency of B7-H1 in CRC tissue of the patients at Duke's stage (C+D) was higher than that of the patients at Duke's stage (A+B)(P<0.01).The positive expression rate of B7-H4 in CRC tissue was higher than that in adjacent normal tissue(χ2=78.92,P<0.01).The differences of expression frequencies of B7-H4 in CRC tissues with different infiltration depths were significant(P<0.05).The high expression frequency of B7-H4 in CRC tissue of the patients with positive lymphnode metastasis was higher than that of the patients with negative lymphnode metastasis(P<0.05).The number of Treg positive expression cells in CRC tissue was significantly higher than that in adjacent normal tissue (P<0.01),the number of Treg postive expression cells in CRC tissues with different differentiation degrees were different(P<0.01),and the number of Treg positive expression cells in CRC tissue of the patients with positive lymphnode metastasis was higher than that of the patients with negative lymph node metastasis(P<0.01).The expression of B7-H1 was correlated with Treg infiltration(P<0.01,Cramer's=0.463);the expression of B7-H4 was correlated with Treg infiltration(P<0.05,Cramer's=0.320). Conclusion: B7-H1 and B7-H4 are highly expressed in human CRC tissue,and they are associated with the increasing of Treg infiltration;they can be used as the indicators to judge the prognosis of CRC.

Influence of porcelain firing and different manufacturing techniques in fit of Co-Cr porcelain crowns

Objective: To explore the influence of different manufacturing techniques and different scanning objects in the marginal and internal fits of the Co-Cr porcelain crowns after procelain firing,and to provide the basis for judging the influencing factors of longevity of restorations. Methods: Five extracted maxillary premolar were selected.According to the prepared tooth,silicone rubber and stone die restoration models,which were fabricated by CAD/CAM and selective laser melting(SLM),the restorations were divided into control group(traditinal casting),tooth-SLM group,silicone rubber-SLM group,stone die-SLM group,tooth-CAD/CAM group,silicone rubber-CAD/CAM group, and stone die-CAD/CAM group.Then all the restorations were fired with the porcelain.The marginal and internal fits of the specinens were measured by micro-compated tomography(Micro-CT). Results: Compared with control group,the AMD of restorations in tooth-SLM and CAD/CAM groups were decresed(P<0.05),and the MG of restorations in silicone rubber-SLM group was decresed(P<0.05).Compared with control group,the CA and AW of restorations in silicone rubber-SLM group were decresed(P<0.05),the AW of restorations in tooth-CAD/CAM group was the least;compared with control group, the AOT and OA of restorations in stone die-SLM group and CAD/CAM group were decresed(P<0.05),and the AOT and OA of restorations in silicone rubber-CAD/CAM were the least.Compared with before porcelain firing,the marginal fit and internal fit of restorations after porcelain firing were incresed(P<0.05).There was positive corrlation between the MG of restorations and fabricated techniques and scanning objects(P<0.05).There was positive corrlation between the average of internal fit of restorations and fabricated techniques and scanning objects(P<0.05). Conclusion: The porcelain firing can affect the marginal and internal fits of Co-Cr porcelain crowns.The restorations fabricated by SLM had a better marginal fit and the restorations fabricated by CAD/CAM has a better internal fit.

Changes of levels of IL-7 and IL-10 in peripheral blood of patients with different clinical stages of eczema and thier clinical significances

Objective: To detect the levels of interleukin-7 (IL-7) and IL-10 in peripheral blood of the patients with different clinical stages of eczama,and to elucidate their clinical significances. Methods: Ninety patients with eczema were selected,and divided into acute eczama group (n=30),subacute ecxama group (n=30),and chronic eczama group (n=30).Meanwhile,30 healthy people who went to hospital for physical examination during the same period were recruited as control group.The relative expression levels of cytokines IL-7 and IL-10 mRNA in peripheral blood of the subjects in various groups were detected by RT-PCR method.The levels of IL-7 and IL-10 in peripheral blood of the subjects in various groups were measured by ELISA method.The eczema area and severity index (EASI) scores of the subjects in various groups were detected.The relationships between IL-7 level, IL-10 level and EASI scores were analyzed. Results: Compared with control group,the relative expression levels IL-7 and IL-10 mRNA in peripheral blood of the patients with different clinical stages of eczenma were significantly increased (F=3.17,P=0.04; F=4.73,P=0.02);the expression levels of IL-7 and IL-10 in peripheral blood of the patients with different clinical stages of eczema were significantly increased (F=6.23,P<0.01; F=5.34,P=0.01).The levels of IL-7 and IL-10 were positively correlated with the corresponding mRNA relative expression levels in peripheral blood of the patients with different clinical stages of eczema (r=0.95,P<0.01; r=0.94,P<0.01) and EASI scores of the patients (r=0.88,P<0.01; r=0.89,P<0.01),which were increased along with the increasing of severity degree of the patients. Conclusion: The levels of IL-7 and IL-10 of the patients with eczema are significantly higher than those in the healthy controls,which are positively correlated with the severity of the disease. The cytokines IL-7 and IL-10 may be involved in the pathological process of eczema,and the detection of their experssion levels contributes to assess the severity of disease.

Analysis on association between single nucleotide polymorphism of selenoprotein S gene and risk of liver cancer

Objective: To investigate the distribution of single nucleotide polymorphism (SNP) of selenoprotein S(SelS)gene in the patients with liver cancer and the healthy controls,and to clarify the correlation of the SNP and the risk of liver cancer in the Inner Mongolia Han population. Methods: The distribution of genotypic and allelic frequencies at rs34713741 site was detected by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)method among 96 patients with liver cancer and 135 healthy controls,and verified by gene sequencing analysis.The relationship between SNP of SelS gene and the risk of liver cancer was analyzed. Results: There were no differences of the frequencies of CC,CT,and TT genotypes and C and T alleles at rs34713741 site of SelS gene between the patients with liver cancer and the healthy controls(χ2=2.679,χ2=2.331,P>0.05).The differences in the frequencies of CC,CT,and TT genotypes and C and T alleles at rs34713741 site of SelS gene of the drinkers between liver cancer group and healthy control group were significant (χ2=6.533,χ2=6.417,P<0.05).The relative risk of liver cancer in the drinkers carrying T allele was 2.297 times of the drinkers carrying genotype CC(OR=2.297,95%CI:1.202-4.393). Conclusion: The gene-environment interaction of SelS and drinking may associate with liver cancer in the Inner Mongolia Han population.

Detection of levels of serum visfatin and PGE2 in patients with periodontitis and their relationships with activity of periodontitis

Objective: To detect the levels of visfatin,prostaglandin E2 (PGE2) and interleukin-1β (IL-1β) in serum of the chronic periodontitis patients,and to investigate the role of the above three factors in the pathogenesis of chronic periodontitis and their clinical significances in the assessment of activity of chronic periodontitis. Methods: A total of 79 cases of chronic periodontitis patients(including active and resting stages) were selected.The chronic periodontitis patients were divided into active stage of chronic periodontitis group (n=33) and resting stage of chronic periodontitis group (n=46) according to the diagnostic criteria for chronic periodontitis activity.A total of 25 cases of healthy volunteers were also selected as healthy control group.The serum of all cases were collected,then the serum levels of visfatin,PGE2 and IL-1β of the subjects in various groups were detected by ELISA method.The relationships between the serum levels of visfatin,PGE2 and IL-1β were analyzed with Spearman and partial correlation analysis. Results: The levels of serum visfatin,PGE2 and IL-1β of the patients in chronic periodontitis group were significantly higher than those in healthy control group(P<0.05).The levels of serum visfatin,PGE2 and IL-1β of the patients in active stage of chronic periodontitis group were significantly higher than those in resting stage of chronic periodontitis group(P<0.05).The correlative analysis results demonstrated that the serum IL-1β level had positive relationships with the serum levels of visfatin and PGE2 of the patients in chronic periodontitis group(r=0.68,P<0.05;r=0.79,P<0.05).The serum visfatin level had positive relationships with the serum PGE2 level of the patients in chronic periodontitis group(r=0.63,P<0.05);the higher the serum visfatin level was,the higher the serum PGE2 level was. The serum visfatin level had significantly positive relationship with the serum PGE2 level of the patients in active stage of chronic periodontitis group(r=0.78,P<0.05).The serum levels of visfatin and PGE2 of the patients in resting stage of chronic periodontitis group had positive correlation at a low level(r=0.45,P<0.05). Conclusion: Visfatin and PGE2 may be involved in the pathogenesis of chronic periodontitis.The increasing of serum visfatin and PGE2 levels may predict the active stage of chronic periodontitis.

Comparsion of GV oocytes transcriptome between PCOS patients and normal ovulatory women

Objective: To investigate the effects of controlled ovarian stimulation(COS) on the differentially expressed genes in GV oocytes and main signal transduction pathways in the polycystic ovary syndrome (PCOS) patients and the normal ovulatory women,and to screen the key genes impacting the development of oocytes of the PCOS patients. Methods: During controll ovarian hyperstimulation with GnRH-a long protocol, 3 patients with PCOS(PCOS group) and 3 normal ovulatory women due to male infertility factor (control group)were selected.Enzyme digestion was used to islolate the granule cells.The dumped immature oocytes were collected after intracytoplasmic sperm injection(ICSI).The cDNA library was constructed and sequencing was performed in Illumina MiSeq sequencing platform and RT-PCR was used to confirm the data obtained in vivo. Results: A total of 510 024 82 sequence reads were obtained,and 8 G base sequence information were contained.A total of 63 differentially expressed genes were found by bio-informatics software,including 19 significant up-regulation genes and 44 significant down-regulation genes (Fold Change>4,FDR<0.01).The expression levels of vascular endothelial growth(VEGF) and fatly acid dehydrogenase 1(FADS1) mRNA in the patients in PCOS group were significantly higher than those in control groups(P<0.05),and the expression levels of Runt-related transcription factor 2(RUNX2),chemokine factor 1 (CXCL1) and heat shock protein27(Hsp27) mRNA were significantly lower than those in control group (P<0.05).These above results were consistent with the functional verification results.According to the GO function annotation,these genes were divided into several branches,including phosphorylation,autophagy,and transcriptional regulation,etc.Referring to the KEGG database,the differential genes were mainly associated with phosphatidylinositol -3-kinase (PI3K-Akt),transforming growth factor β(TGF-β) Hippo,p53 and peroxisome proliferators-activated receptor(PPAR) signal pathways. Conclusion: The differentially expressed genes in GV oocytes of the PCOS patients may affect the development of follicle and maturation of oocytes and lead to poor quality of embryo.

Objective: To explore the expressions of interleukin-17E(IL-17E),interleukin -17F(IL-17F) and their receptors interleukin -17RB(IL-17RB),interleukin -17RC(IL-17RC) in enteritis,intestinal polyps and colorectal carcinoma tissues,and to analyze their relationships with the malignancy of colorectal carcinoma. Methods: Immunohistochemistry staining was used to detect the expressions of IL-17E,IL-17RB,IL-17F, and IL-17RC in the tissues of enteritis (n=15),intestinal polyps (n=5) and colorectal carcinoma (n=30).The relationships between IL-17E and IL-17RB,IL-17F and IL-17RC,and the relationships between IL-17E,IL-17F and malignancy of colorectal carcinoma were analyzed. Results: IL-17E,IL-17F,IL-17RB and IL-17RC mainly expressed in glandular epithelial cells,mononuclear cells and vascular endothelial cells as well as partial carcinoma cells.The positive expression rates of IL-17E in enteritis and intestinal polyps tissues were higher than that in colorectal carcinoma tissue (P<0.05).Compared with enteritis tissue,the positive expression rates of IL-17F in intestinal polyps and colorectal carcinoma tissues were increased (P<0.05),which was increased with the increasing of malignancy of coloretal carcinoma.Compared with colorectal carcinoma tissue,the positive expression rates of IL-17RB in enteritis and intestinal polyps tissues were increased (P<0.05).Compared with enteritis and intestinal polyps tissues,the positive expression rate of IL-17RC in colorectal carcinoma tissue was increased (P<0.05).There was positive correlation between the positive expression rate of IL-17F and the positive expression rate of IL-17RC in colorectal carcinoma tissue(r=0.667,P=0.001). Conclusion: IL-17F,IL-17RC,IL-17E,and IL-17RB signals may take part in the incidence and progression of colorectal carcinoma.

Analysis on functional status of major joints in limbs of moderate hemophilia patients

Objective: To detect the functional status of major joints in limbs of the patients with moderate hemophilia,and to improve the understanding and management of hemophilia. Methods: A total of 93 patients with moderate hemophilia (FVⅢ:C/FⅨ:C:1%-5%) were divided into seven subgroups according to their ages:<10 years,10-20 years,20-30 years,30-40 years,40-50 years,50-60 years,and > 60 years. The joint involvement rates of patients were observed and the joint function was evaluated using Hemophilia Joint Heath Score (HJHS). Results: Compared with moderate hemophilia group,the difference in the involvement rate of hemophilic arthritis of the patients in severe hemophilia group was not significant(P>0.05).The mean HJHS of the patients in moderate hemophilia group and severe hemophilia group had no statistical difference (P=0.157).In 20-30 years group,the mean HJHS of the patients in severe hemophilia group was higher than that in moderate hemophilia group(Z=-2.127,P=0.033). In 10-20 years and 20-30 years groups,there were statistically significant differences in the HJHS of knee joint between moderate hemophilia group and severe hemophilia group(Z10-20=-2.710,P10-20=0.007;Z20-30=-2.384,P20-30=0.017).There were no significant differences in the HJHS of the patients in moderate hemophilia group and severe hemophilia group at the different ages (P>0.05). Conclusion: The hemorrhage treatment status of moderate hemophilia patients is poor.The combination of on-demand treatment and preventive treatment can reduce the joint hemorrhage,and guarantee the joint function of hemophilia patients effectively and improve the life quality of the patients.

Objective: To compare the changes of soft and hard tissue profiles of the adolescent patients with class Ⅱ division 1 malocclusion between before and after treated with two different kinds of tooth extraction methods, and to provide the reference for clinical treatment. Methods: A total of 60 adolescent patients with class Ⅱ division 1 malocclusion were included,and they were divided into 44/44 group(n=30) and 44/55 group(n=30) based on different kinds of tooth extraction methods. Four first premolars of the patients in 44/44 group were extracted and 2 maxillary first premolars and 2 mandibular second premolars of the patients in 44/55 group were extracted.The measurement indexes of pre-treatment(T1) and post-treatment(T2) and their difference values(T2-T1) of soft and hard tissues of the adolescent patients were detected by lateral cephalometric radiographs. Results: As for the hard tissue,compared with before treatment,the Overjet,SNA,ANB,U1-NA,distance of U1-NA,U1-SN,NA-PA,and A-NP of the adolescent patients in two groups after treatment were decreasd (P<0.01),and U1-L1 and Pog-NB were increased (P<0.05 or P<0.01); as for the soft tissue, compared with before treatment, the H angle, UL-SnPg', LL-SnPg', UL-ELine, LL-ELine, and Sn-HLine of the adolescent patients in two groups after treatment were decreased(P<0.05 or P<0.01); B'LL-FH was increased(P<0.01);there were no significant differences in the difference values(T2-T1) of measurement indexes in the soft and hard tissues of the adolescent patients in two groups (P>0.05). Conclusion: The changes of soft and hard tissue profiles of the adolescent patients after treated with two different kinds of tooth extraction methods are not significant.

Evaluation on clinical application of Florida probes in probing peri-implantsoft tissue

Objective: To detect the peri-implant soft tissue with Florida probes,and to provide the clinical basis for choosing the appropriate clinical examination method,probing strength,and treatment of peri-implant soft tissue in the maintenance period. Methods: The common periodontal probes and Florida probes were used to examine the probing depth(PD) values in 62 patients who underwent implantation for more than 6 months for two times,and the contralateral teeth were natural teeth.The natural teeth and the implants were divided into inflammation group(n=32) and healthy group(n=30) according to whether the bleeding was probed.The coefficient of variation (CV) values of PD values of natural teeth and peri-implant soft tissues detected by two probes were compared. Results: There were no statistically significant differences in the PD values between natural teeth and implants of the patients measured by common periodontal probes in healthy and inflammation groups (P>0.05).There were no statistically significant differences in the PD values between natural teeth and implants of the patients measured by Florida probes in healthy and inflammation groups (P>0.05).The CV value of PD value detected by Florida probes of the patients in healthy group was less than that detected by common periodontal probes (t=2.489,P=0.019); the CV value of PD value detected by Florida probes of the patients in inflammation group was less than that detected by common periodontal probe (t=2.238,P=0.033).The CV value of PD value of implants of the patients in inflammation group was higher than that in healthy group(Z=3.804,P<0.01;Z=2.913,P=0.004);the CV value of PD value of nature teeth of the patients in two groups had no significant differences(P>0.05). Conclusion: Both two probes have good reproducibility in probing the healthy and inflammatory sites of the natural teeth and implants.The Florida probes are more reproducible than the common periodontal probes in probing the healthy and inflammatory peri-implant soft tissues.

Evaluation on efficacy of Huaiqihuang granules in treatment of children with asthma at non-acute attack stage

Objective: To explore the efficacy of Huaiqihuang Granules in the treatment of the children with asthma at non-acute attack stage and its effect on the immune function indexes of body. Methods: A total of 180 children with asthma at non-acute attack stage were divided into Huaiqihuang Granules treatment group (n=92,treated with traditional Chinese medicine Huaiqihuang Granules along with oral montelukast and inhaled corticosteroid,ICS+MK+Hgroup) and routine treatment group(n=88,only given oral montelukast and inhaled corticosteroid,ICS+MK group).The indexes of asthma attack degrees,traditional Chinese medicine (TCM) syndrome indexes, blood immune function indexes and pulmonary function indexes of the children in two groups were detected. Results: There was no difference in the disease degrees of the children between two groups before treatment(P>0.05).Compared with routine treatment group,the score of acute attack of asthma of the children in Huaihuang Granules treatment group was decreased after treatment(P<0.05).Compared with routine treatment group,the scores of dry/loose stools,low-grade fever,cough/expectoration, spiritlessness/anorexia and white tongue fur of the children in Huaiqihuang Granules treatment group were significantly decreased (P<0.01),and the scores of perspire during sleep and dry mouth with desire for drinking were decreased(P<0.05).Compared with routine treatment group,the levels of immunoglobulins (serum lgA,lgG and lgM) of the children in Huaiqihuang Granules treatment group were increased after treatment(P<0.05)Compared with routine treatment group,the percentages of T cells,Th cells and NK cells of the children in Huaiqihuang Granules treatment group were increased(P<0.05),the percentage of B cells was decreased(P<0.05),and there was no difference in the percentages of serum Ts cells of the children between two groups(P>0.05).Compared with routine treatment group,the peak expiratory flow (PEF) and the ratio value of forced expiratory volume in the first second/forced vital capacity(FEV1/FVC) of the children in Huaiqihuang Granules treatment group were increased (P<0.05),but there was no difference in FEV1 of the children between two groups(P>0.05). Conclusion: On the basis of routine treatment,Huaiqihuang Granules can reduce the condition of children with asthma at non-acute attack stage and improve the TCM syndrome and immune function.

Objective: To detect the clinical effect of Twin-Block appliance in the treatment of the patients with Class Ⅱ division 1 malocclusion,and to provide the basis for clinical treatment. Methods: A total of 30 patients with class Ⅱ division 1 malocclusion were treated with Twin-Block appliance.The variables of the patients before and after treatment were measured with Ricketts cephalometric radiographys. Results: Compared with before treatment, the length of corpora mandibulae(Xi-Pm) of the patients after treatment was increased by 2.50 mm (P<0.05),the SNB angle was increased by 2.58°(P<0.05),and the ANB was decreased by 2.71°(P<0.01);the molar relationship distance and the Overjet were decreased by 2.81 mm and 3.68 mm,respectively (P<0.01);the L1-OP was decreased by 1.89 mm,the distance of L1-APog was increased by 1.84 mm, and the angle U1-APog was decreased by 7.35°(P<0.01);the Ls-TVL was increased by 1.85 mm,the Sm-TVL was increased by 2.12 mm (P<0.05),and the Pos-TVL was increased by 2.89 mm (P<0.05). Conclusion: The Twin-Block appliance could obviously promote the development of mandibular bone,upright the anterior teeth and improve the lip angle of upper anterior teeth and the proflie in the treatment of the patients with class Ⅱ division 1 malocclusion in the growth development period.

Clinical application of modified transthecal digital block in treatment of handtrauma in children

Objective: To probe the advantages and clinical application value of modified transthecal digital block in the treatment of hand trauma in the children,and to provide the references for the selection of anesthesia methods for surgery. Methods: A total of 102 patients (104 digits) required acute or chronic hand surgery were selected.The patients were divided into two groups by the method of random number table,54 patients(54 fingers) were treated with traditional digital block(traditional digital block group),and 48 patients (50 fingers) were anesthetized with modified transthecal digital block(modified transthecal digital block group).The pain detection was performed on the 4 sites within 5 min after anesthesia by using the method of Face,Legs,Activity,Cry,Consolability Behavioral Tool(FLACC)(<7 years old) or Visual Analogue Scale(VAS)(7-18 years old).The 0 score of pain meant that the anesthesia was successful in 5 min after injection.After 5 min,if the child still felt painful and unable to undergo surgery,it was marked as failure,and the method of traditional digital block was used to complete the operation.The successful rate,the onset time,the duration time,and the dose of anesthetic drugs of the patients in two groups were detected. Results: The successful rate of the patients in modified transthecal digital block group had no significantly difference compared with traditional digital block group (P>0.05).Compared with traditional digital block group,the average onset time of anesthetic drug of the patients in modified transthecal digital block group was shortened (P<0.05),and the average dose of anesthetic drug was decreased (P<0.05). Conclusion: The modified transthecal digital block has the advantages of smaller dose,quicker effect,easier operation,and less pain.It is suitable for the clinical application in the treatment of hand trauma in the pediatric patients.

Objective: To investigate the clinical effect and treatment strategy of percutaneous endoscopic interlaminar discectomy(PEID) in the treatment of calcified L5S1 lumbar disc herniation. Methods: A total of 15 patients with calcified L5S1 lumbar disc herniation were selected and treated with PEID combined with variable power system.The Visual Analogue scale (VAS) and Oswestry dysfunction index (ODI) scores of the patients were evaluated before operation,and 1 d,1 week,3 months,6 months after operation.Macnab score was used to evaluate the curative effect of the patients 6 months after operation. Results: The scores of VAS and ODI of the patients 1 d,1 week,3 months, and 6 months after operation were significantly lower than those before operation (P<0.05);the modified Macnab score showed that the excellent and good rate was 86.67%;the postoperative image results showed that the calcification area of intervertebral disc was removed,the nerve root was decompressed effectively,and there were no nerve root injury,cerebrospinal fluid leakage and infection. Conclusion: PEID combined with variable power system can effectively treat the calcified L5S1 lumbar disc herniation with the advantages of less trauma,short operation time and short recovery period.

Objective: To explore the clinical efficacy of digital arteria axial flap augment volume in the treatment of Wassel type Ⅲ-Ⅴ congentital thumb duplication,and to provide the basis for its clinical treatment. Methods: The thumb augment volume surgery was performed in 14 children with Wassel type Ⅲ-Ⅴ congenital thumb dupliation.A narrow and long flap with vascular vessel and soft tissue was designed with microsurgical technique to augment the soft tissue of the reconstructed thumb and reconstruct the perimeter and appearances of the reconstructed thumb.The perimeters and appearences of the reconstructed thumb and healthy thumb were detected before and after surgery. Results: All the flaps were survived after the surgery.The average perimeter of the reconstructed thumb was 89%-104% relative to the healthy contralateral thumb based on the data detected from the tuberositas unguicularis,proximal interphalangeal joint,proximal phalanx midshaft, metacarpophalangeal joint and metacarpal midshaft.The appearance of reconstructed thumb was more similar to that of healthy thumb. Conclusion: The digital arteria axial vascular flaps which are used to augment the volume of reconstructed thumb of Wassel type Ⅲ-Ⅴ congenital thumb duplication can be easily obtained and the reconstructed thumbs have better appearences.The surgery is an effective way to improve the thin and small appearences of reconstructed thumbs.

Treatment of lung squamous cell carcinoma with apatinib in patient who couldn't tolerate chemotherapy: A case report and literature review

Objective: To analyze the efficacy of apatinibin the treatment of a patient with lung squamous cell carcinoma who couldn't tolerare the chemotherapy and review the literature,and to clarify the effectiveness of apatinib in order to provide the treatment reference. Methods: The patient was clearly diagnosed as stage Ⅳlung squamous cell carcinoma.After four cycles of chemotherapy,there was no significant change of the tumor volume,while the patient couldn't tolerate the side effects caused by chemotherapy.Then the disease progressed rapidly.The primary lesion and multiple metastases responded significantly to the radiotherapy.However,the lesions were too extensive to radiotherapy,therefore,apatinib(500 mg·d-1,40 d) was used to inhibit the progression of disease. Results: After the application of apatinib,the volumes of both lesions in chest wall and lung shrank greatly compared wtih before administration.The patient developed myelosuppression(grade Ⅲ thrombocytopenia) and stopped taking apatinib.Then the volume of lesion in chest wall was increased.The dosage of apatinib reduced to 250 mg·d-1,and the progression free survival(PFS) of the patient reached to 5 months. Conclusion: For the patients with lung squamous cell carcinoma who couldn't tolerate chemotherapy or fail in multiple lines of chemotherapy,apatinib has better efficacy with controllable side effects.

Epidural hematoma ossification after craniocerebral injury: A case report and literature review

Objective: To analyze the clinical data of a patient with epidural hematoma ossification after craniocerebral injury,and to improve the understanding and treatment level of the disease by combing the literature review. Methods: A 36-year-old man with headache and dizziness for 3 months was performed general physical examination and related imaging examination to identify the location and size of the lesion and its relationships with skull and endocranium.The patient with multiple myeloma was treated with chemotherapy for 4 months.The changes of imaging features of the intracranial lesions were judged.The diagnosis was confirmed by biopsy pathology result after operation and the patient was given the corresponding treatment. Results: The diagnosis was regarded as epidural hematoma ossification after craniocerebral injury according to the patient's imaging examination and the pathological results after operation.The diagnosis results of skull osteoma and multiple myeloma metastasis were excluded.The lesion was stable during the follow-up of two years. Conclusion: For the patient with epidural hematoma ossification after craniocerebral injury,the skull osteoma should be considered,and other bone diseases should be excluded.The imaging evidence is needed for the diagnosis.The results of biopsy pathology should be taken if necessary.

Solid pseudopapillary neoplasm of pancreas arising from majus omentum: A case report and literature review

Objective: To explore the pathological diagnosis features and differential diagnosis of one case of patient with solid pseudopapillary neoplasm (SPN) of pancreas occurred in the majus omentum,and to improve the understanding of extra-pancreatic SPN. Methods: One female patient was admitted to hospital because of abdominal mass found 20 d ago.The patient was diagnosed as multiple hysteromyoma by gynecological color ultrasound,and the results of abdominal ultrasound had no obvious abnormity.The patient underwent hysterotomy,and a well-defined mass in the majus omentum was found during operation,so the patient received hysterectomy and omental tumor resection. Results: The patient was confirmed as multiple hysteromyoma by the postoperative pathological examination.The patient was clearly diagnosed as SPN arising from the majus omentum on the basis of pathological characteristics and immunohistochemical staining results.There were no ectopic pancreas tissue arround the tumor and no SNP metastasis in examination of ultrasound and CT.No recurrence and metastasis were found in the follow-up of 3 years. Conclusion: SPN arising from the majus omentum is extremely rare.The tumor has clear boundary,typical morphology and immunohistochemical appearances.

Rare huge serous cystadenoma of broad ligament: A case report and literature review

Objective: To analyze the clinical data and surgical treatment procedures of a patient with rare huge serous cystadenoma of broad ligament,to clarify its diagnostic and treatment characteristics,and to explore the present situation and advantages of laparoscope in the operation of broad ligament tumor. Methods: The patient was admitted to hospital due to abdominal distension for 2 years and aggravated illness for 2 months.The diagnosis result was pelvic and abdominal mass arising from ovarian according to the preoperative color Doppler ultrasound and physical examination.Based on the laparotomy and intraoperative rapid pathological results,the patient was diagnosed as huge serous cystadenoma of broad ligament.The location of ureter and uteri artery and vein were carefully identified during the operation and they were ligated for hemostasis at the basal part of tumor.The operation was successful. Results: The ultrasonography image of the huge serous cystadenoma of broad ligament was very similar to that of large ovarian tumor.The misdiagnosis rate of ultrasound was high,and CT and MRI were important for the identification of large broad ligament tumor;the accuracy rate of preoperative diagnosis could be improved by CT and MRI.Carefully blunt separation close to the tumor surface during the operation could avoid the ureteral injury.The patient recovered well and discharged from hospital. Conclusion: The symptoms of huge serous cystadenoma of broad ligament patient are lack of specificity.The accuracy rate of preoperative diagnosis should be improved in combination with the medical history and imageological findings of the patient.The operation plan should be chosen reasonably and the operation should be carefully performed to avoid the occurrence of secondary injuries.

Objective: To explore the optimal cutoff values of blood pressure-to-height ratio (BPHR) for screening the hypertension of the adolescents,and to provide the basis for understanding the prevalence of hypertension in the adolescents in Yanji city of Jilin province. Methods: From September to October 2016,a total of 23 middle schools and high schools in Yanji city of Jilin province were selected,and a total of 18 244 adolescents (9 567 boys and 8 677 girls) aged from 12 to 17 years old in Yanji city of Jilin province were enrolled by means of whole population sampling.The blood pressure (BP),heights and body weights of the adolescents were measured,and the systolic BP-to-height ratio (SBPHR) and diastolic BP-to-height ratio (DBPHR) were calculated.Receiver operating characteristic (ROC) curve analysis was performed to determine the optimal cutoff values and evaluate the sensitivity,specificity,negative predictive value (NPV) and positive predictive value (PPV). Results: The prevalence of hypertension in the Korean and Han adolescents aged 12-17 years old were 12.8% and 13.0% for boys and 5.9% and 6.2% for girls,respectively; there was no significant difference in the prevalence of hypertension between different genders in two nationalities (χ2=0.091,P> 0.05;χ2=0.295,P> 0.05).The total prevalence of hypertension was 12.9% for boys and 6.1% for girls,the difference was statistically significant (χ2=242.072,P<0.01). The prevalence of hypertension of the boys at the age of 15 years old reached its peak and the curve of hypertension prevalence changed with age showed as a sudden increasing and sudden decreasing trend;the changes of prevalence of hypertension with age were little in the girls.The prevalence of hypertension in the boys aged 12 to 17 years old was significantly higher than that in the girls (P<0.05).The correlation between age and SBPHR in the boys was negative (r=-0.085,P<0.01),and there was no correlation between age and SBPHR in the girls(r=0.018,P>0.05).The correlation between age and DBPHR in the boys aged 12-17 years old was negative (r=-0.080,P<0.01),and there was no correlation in the girls(r=-0.017, P>0.05);all the correlation coefficients were less than 0.1,so it was judged that there was no significant correlation between age and BPHR.The threshold values of SBP screened by SBPHR were 0.73 mmHg·cm-1 in the boys aged 12-17 years old (sensitivity:92.2%,specificity:88.9%,PPV:52.6%,NPV:98.8%) and 0.75 mmHg·cm-1 in the girls(sensitivity:95.6%,specificity:89.5%,PPV:33.6%,NPV:99.6%),respectively. The threshold values of DBP screened by DBPHR were 0.49 mmHg·cm-1 in the boys aged 12-17 years old (sensitivity:91.6%,specificity:93.9%,PPV:35.3%,NPV:99.6%) and 0.51 mmHg·cm-1 in the girls (sensitivity:89.8%,specificity:94.3%,PPV:26.7%,NPV:99.8%),respectively. Conclusion: The prevalence of hypertension in the adolescents aged 12-17 years old in Yanji city of Jilin province is lower than the national average value,and BPHR can be used as an index to screen the hypertension of the adolescents aged 12-17 years old in Yanji city of Jilin province.

Analysis on risk factors related to incidence of patientswithcolorectal polyps

Objective: To investigate the effects of gender,age,and blood lipid levels on the incidence of colorectal polyps,and to clarify the risk factors of colorectal polyps,and to provide the reference for clinical diagnosis and treatment of colorectal polyps. Methods: The serum lipid levels of the patients with colorectal polyps (polyps group,n=239) and the healthy controls or the patients with chronic nonspecific enteritis (control group,n=245) were examined.The effects of gender, age and blood lipid level on the incidence of colorectal polyps were analyzed,and the levels of blood lipids of the colorectal polyps patients with different pathogenesis parts and pathological types were detected and compared. Results: The incidence of colorectal polyps of the patients with high level of low density lipoprotein cholesterol(LDL-C) (P<0.01,OR=6.26,95%CI:3.79-10.33) and high level of triglyceride(TG)(P=0.043,OR=1.63,95%CI:1.02-2.63) and male (P=0.001,OR=2.05,95%CI:1.35-3.11) and the patients whose ages were older than 66 years old were increased. There were no significant differences in the levels of high density lipoprotein cholesterol(HDL-C) and total cholesterol (TC) of the patients between two groups (P>0.05).The levels of LDL-C(t=-4.497,P<0.01),TG (t=-2.116,P=0.036),TC (t=-3.843,P<0.01) and ratio of hyperlipidaemia(HLP) (χ2=9.458,P=0.002)in the patients with left colorectal polyps were higher than those in the patients with right colorectal polyps;but there were no significant differences in the gender,age and HDL-C level of the patients in these two groups (P>0.05).The level of LDL-C of the patients with non-adenomatous colorectal polyps was higher than that of the patients with adenomatous colorectal polyps (t=2.049,P=0.042);but there were no statistically significances in the gender,age and HDL-C level,TG level,TC level, and HLP ratio of the patients between two groups(P>0.05). Conclusion: Male,older,high levels of LDL-C and TG are the independent risk factors for colorectal polyps.High level of LDL-C is associated with non-adenomatous colorectal polyps.

Survey analysis on birth defects among children aged 0-5 years old in Jilin province

Objective: To investigate the situation and distribution characteristics of birh defects among the children aged 0-5 years old in Jilin province,and to discuss the intervention countermeasures of birth defects. Methods: The subjects were selected through multi-stage proportional cluster random sampling method.In the eastern,central and western regions of Jilin province,a total of 90 townships (towns,streets) in 15 counties (cities,districts) were selected as the survey sites.A retrospective study was conducted on all the children born from 1 October 2008 to 30 September 2013 in all the survey sites.A total of 48 720 samples were collected from 0-5 years old children in Jilin province,291 samples were excluded,and 48 429 were valid samples.The self-designed questionnaire was used for survey, Epi Data 3.1 software was used for the entry of data,SPSS 21.0 statistical software was used for the statistical analysis.χ2 test was used to compare the incidence of birth defects of the children. Results: There were 48 429 effective samples and 517 cases of them were defected children.The total incidence of birth defects of the children aged 0-5 years in Jilin province was 10.68 ‰. The incidence of boys(11.52‰) was higher than that of girls(9.56‰)(χ2=83.768,P<0.01).The incidence of birth defects of the children in countryside(11.87‰) was higher than that in town(9.64‰)(χ2=5.651,P=0.017).The top five kinds of diseases with high incidence of birth defects were congenital heart disease,polydactyly (toe),congenital hydrocephalus,cleft lip and cleft palate,and spina bifida. Conclusion: The incidence of birth defects of the children aged 0-5 years old in Jilin province is at a low level,and the differences in the incidence of birth defects between genders and urban and rural is significant. Congenital heart disease has the highest detection rate in birth defects in Jilin province.

Association between salted meat consumption and esophageal squamous cell carcinoma of population in high risk area of esophageal squamous cell carcinoma

Objective: To analyze the levels of dietary intake among the normal population,the esophagitis patients,and the esophageal squamous cell carcinoma(ESCC)patients,and to elucidate the relationship between the diet and the incidence of ESCC. Methods: Frequency-matched case-control study was performed.The normal esophagus population(normal group),the esophagitis patients(esophagitis group) and the ESCC patients(ESCC group) (n=72) were enrolled.The general informations,intake conditions and relationship between the intake and ESCC of the subjects among three groups were compared.Unconditional Logistic regression model was used to estimate the OR and 95%CI when salted meat consumption or not,frenquency of salted meat consumption and time of salted meat consumption in 1 year were brought into the analysis. Results: The family cancer history and family ESCC history in first-degree relatives of the subjects among three groups had significant differences(P<0.01).Compared with normal group and esophagitis group,the ESCC patients got less fruit intake (P<0.01),more picked vegetable(P=0.003) and corn intake(P<0.01).The frenquency of salted meat consumption of the patients in ESCC group was higher than that in normal group (P=0.032).Compared with normal group, the risk of ESCC of the subjects whose consumption time of salted meat was 3-6 months or ≥ 6 months per year was increased(OR=5.78,95%CI:1.36-24.64;OR=6.87,95%CI:1.25-37.87), respectively.Compared with esophagitis group,the risk of ESCC of the subjects whose consumption frequency of salted meat was ≥ 1 time per week was increased (OR=6.48,95%CI:1.65-25.49);the risk of ESCC of the subjects whose consumption time of salted meat was 3-6 months or ≥ 6 months per year was increased(OR=8.70,95%CI:2.07-36.55;OR=44.38,95%CI:5.34-368.65). Conclusion: For the normal population and the esophagitis patients,the excessive consumption of salted meat maybe increase the risk of ESCC.Healthy eating habits may reduce the risk of ESCC.

Objective: To purify the recombinant protein by using the constructed pET28a-ΔPTP-oc recombinant expression plasmid,and to prepare the rabbit polyclonal antibody against osteoclastic protein tyrosine phosphatase(PTP-oc) and to identify its properties. Methods: The constructed pET28a-ΔPTP-oc recombinant plasmid was transformed into E.coli BL21(DE3),and the isopropyl-β-D-thiogalactoside (IPTG) was used to induce the expression of recombinant protein.The recombinant protein was purified by the method of Q fast flow agarose and Ni-NTA agarose,and the purified recombinant protein was used as an antigen to immunize the rabbit to obtain PTP-oc polyclonal antibody.The titer and specificity of the antibody were identified by indirect ELISA and Western blotting methods. Results: The recombinant PTP-oc protein was successfully purified.The indirect ELISA results showed that after immunizing the rabbit with the recombinant PTP-oc protein,the polyclonal antibody with the titer of 1:3 2000 was gotten.The Western blotting results showed that the polyclonal antibody had high specificity. Conclusion: The PTP-oc recombinant protein is successfully purified,and the polyclonal antibody of PTP-oc is prepared successfully.
