光活化的金丝桃素对人前列腺癌细胞PC3M的生长抑制作用

摘要: 目的:探讨光活化的金丝桃素对人前列腺癌细胞PC3M的抑癌作用。 方法:用生长抑制试验和流式细胞技术观察给予金丝桃素后PC3M细胞的增殖、细胞周期和凋亡的变化。 结果:不同浓度的金丝桃素在体外可显著抑制PC3M细胞增殖(P<0.05),药物浓度为0.5、1.0和2.0 mg•L-1时,抑制率分别为21%、50%和91%;细胞周期出现S期阻滞,由S期进入M2期的比例为零,并诱导PC3M细胞凋亡。 结论:金丝桃素可抑制前列腺癌细胞增殖并诱导细胞凋亡。

Abstract: Objective To study the influence of hypericin on the proliferation and apoptosis of cultured human prostate cancer cells. Methods Growth inhibition test and flow cytometry were used to examine the changes of proliferation, cell cycle and apoptosis. Results Hypericin of different concentrations could significantly inhibit the proliferation and induce the apoptosis of human prostate cancer cells cultured in vitro. When the concentrations were 0.5,1.0,2.0 mg•L-1,the inhibitory rates were 21%,50%, and 91%,respectively. S phase was blocked and didn′t change into M2 phase. Conclusion Hypericin can inhibit the proliferation and cell cycle of cultured human prostate cancer cell and induce its apoptosis.
