Effects of BPI on expressions of TNF-α and IL-6 in middleear mucosa of rats with otitis media with effusion

摘要: 目的:探讨杀菌-通透性增强蛋白(BPI)对分泌性中耳炎(OME)大鼠中耳黏膜TNF-α和IL-6 mRNA及蛋白表达的影响和意义。 方法:采用大鼠中耳腔注射脂多糖(Lipopolysaccharide,LPS)制备OME模型,动物分为LPS实验组和BPI治疗组。利用ELISA和RT-PCR方法检测中耳黏膜组织TNF-α和IL-6 mRNA及其蛋白的表达。结果:LPS实验组和BPI治疗组在第1、2天均有IL-6的表达,但LPS组表达量高于BPI组(P<0.05)。在第7天后两组均无表达;而TNF-α的表达贯穿于两组实验的始终,但LPS组表达量高于BPI组(P<0.05)。结论:IL-6和TNF-α在中耳炎病变的形成中起作用;BPI通过抑制细胞因子表达和分泌,减轻炎症刺激;BPI是一个潜在的治疗OME 的有效药物,可能预防慢性OME的发生。

Abstract: Objective To study the effect and significance of bactericidal/permeability increasing protein (BPI) on the TNF-α and IL-6 mRNA expressions and protein levels in the middle ear mucosa of the rat models of otitis media with effusion (OME). Methods Rats were subjected to OME by injecting lipopolysaccharide (LPS) into middle ear cavity, and animals were divided into LPS group and BPI group. TNF-α and IL-6 mRNA expressions and their protein levels in middle ear mucosa were measured at various intervals with ELISA and RT-PCR. Results IL-6 mRNA expressions and protein levels peaked at the 1st and 2nd day in LPS group and BPI group. However, IL-6 mRNA in BPI group as well as protein levels were significantly reduced compared with LPS group(P<0.05).There were no expressions of IL-6 mRNA and protein in the LPS group and BPI can group after 7 days. However, TNF-α mRNA expressions and protein levels persisted in LPS group and BPI group. TNF-α mRNA in BPI group as well as protein levels were significantly reduced compared with LPS group(P<0.05). Conclusion IL-6 and TNF-α have an important role in the development of OME; BPI can prevent the expression and secretion of cytokines, reduce inflammatory activities of cytokines; BPI is a potential drug for preventing the occurrence of chronic OME.
