P-糖蛋白基因疫苗诱导小鼠实验性肝炎

Experimental hepatitis induced by P-glycoproteingene vaccine in mice

摘要: 目的:观察P-糖蛋白基因疫苗免疫小鼠诱导的免疫病理变化。方法:P-糖蛋白基因疫苗20~100 μg两次肌注(间隔9 d)免疫小鼠,收集血清检测特异性抗体效价;取心、肝、肾制作组织切片,HE染色行病理检查。结果:末次免疫后5 d鼠血清特异性抗体效价达到 1∶4 000;心、肾组织正常,但肝脏出现自身免疫性肝炎改变。 结论:P-糖蛋白基因疫苗免疫小鼠可诱导实验性肝炎,实验性肝炎小鼠可作为人自身免疫性肝炎的动物模型。

Abstract: Objective To investigate the immunopathological change of the mice immunized with P-glycoprotein gene vaccine. Methods The mice were twicely immunized with either P-glycoprotein gene vaccine at 20-100 μg or the control vector at a 9 days interval. The murine sera were collected to analyze antibody (Ab) titer. Tissue samples of the heart,liver and kidney of the mice were examined by HE staining. Results The titer of Pgp-specific IgG Ab reached as high as 1∶4 000. Pathological damage was found in liver of the immunized mice, indicated by infiltration of mononuclear cells and scattered necrosis of the hepatocytes. Conclusion The P-glycoprotein gene vaccine can induce the experimental hepatitis in mice, which may be used as an animal model for human autoimmune hepatitis.
